Abstract: 

Aim: In this study the effect of AlCl3 toxicity on apoptosis in cells suspension of two rice cultivars Khazar, Tarom were investigated.Material and Methods: In present research cells suspension of two rice cultivars (Khazar and Tarom) grew up in liquid Murashig and Skoog medium within a period of 3 weeks. Then cells were treated with different concentrations (0, 40, 80 and 120 mmolL-1) of AlCl3 for 56 hours. Biochemical changes of DNA by gel electrophoresis and TUNEL test and morphological changes by Hoechst and propidium iodide (PI) staining were investigated.Results: Results of gel electrophoresis and TUNEL test showed DNA damage in these cultivars. Results of Hoechst and PI dying also showed morphological changes such as: condensation of nucleus, membrane damage and shrinkage of cytoplasm.Conclusion: AlCl3 induced nuclear DNA damage and also morphological change in two cultivars of rice especially Tarom by increasing concentration after 56 hours of treats.

Abstract: 

Aim: In this study the effect of vitreous humor on the mesenchymal stem cells (MSCs) derived from bone marrow, adipose tissue and amniotic fluid to lens like cells was investigated.Material and methods: Inthis experimental study collected stem cells from femurs bone marrow and inguinal fat pads of mature NMRI mice and amniotic fluid of 13 days embryos of NMRI mice were cultured. The mesenchymal character of these cells was proven by flow cytometry markers like CD90, CD31. This experiment took place for 14 days with different dosages 10, 15, 20, 30, 40, 50 % of bovine vitreous humor. Express of crystalline markers were detected by immunocytochemistry in experimental and control groups.Results: The flow cytometeric analyses of surface markers were shown expression of CD90 by bone marrow and amniotic fluid stem cells and adipose tissue stem cells (29.29%, 0.57%, 82%, respectively). Moreover, mesenchymal stem cells from these 3 sources had expression of CD31 (3.44%, 1.53%, 0.1%, respectively). Immunocytochemistry results revealed that 40 % vitreous humor in culture media fluid more inducing effect on adipose and amniotic fluid mesenchymal stem cells and 15 % vitreous humor on bone marrow stem cells in order to differentiate.Conclusion: According to the findings of this study, it can be concluded that MSCs derived from all sources can differentiate into lens fiber like cells by inducing effect of vitreous humor.

Abstract: 

Aim: In this study, with the aim of optimizing hairy root growth conditions, leaf, hypocotyl and root explants were inoculated by strain ‘A13’ of Agrobacterium rhizogenes. Then effect of four basal MS liquid media was considered with different combinations and temperature on rate of hairy roots growth.Material and methods: In order to hairy root induction via spary and leaf disc methods, two strains of Agrobacterium rhizogenes (A13 and 9534) were used. For confirmation of transformation but not infection of hairy roots, PCR reaction was performed through specific amplification of rolB and virD genes. Obtained hairy roots in a basal liquid MS medium were cultured under different treatments in based on completely randomized design (CRD) with three replicates and dry weight of hairy roots was measured after two months.Results: Hairy roots were observed in root, hypocotyl and leaf explants after 8 days. The highest percentage of transformation was found in leaf explants. The obtained results showed that the highest (0.19 gr) and the lowest (0.05 gr) rate of hairy root dry weight related to basal liquid MS medium containing 30 g/l sucrose in 35oC and basal MS liquid medium containing 0.2 gr/l NAA hormone, respectively,.Conclusion: The results indicate the main role of culture medium compounds and temperature in enhancing of hairy root biomass.

Abstract: 

Aim: The aim of present study was to demonstrate that the obstruction of CSF flow in hydrocephalic Texas (HT-x) rat and the resulting changes in its protein components can cause abnormal cortical development.Material and Methods: Cortical cells were taken from E18 and E21 Wistar fetuses and cultured for 24 hours in Neurobasal medium. Two groups of experiments were explored. These cells were treated with heated and non-heated CSF collected from E20 and E21fetuses. The cells were cultured for a further 48 hours.At the end of this time, proliferation assays and morphological analysis were performed.Results: Prenatal affected CSF had an inhibitory effect on the proliferation of normal primary fetal cortical cells. Heating the CSF removed this effect.Conclusion: CSF contains important factors that can stimulate both proliferation and cell differentiation and that some of these are likely to be proteins. Disruption of CSF flow can cause the cortical deficiencies in patients with hydrocephalous.

Abstract: 

Aim: The aim of this study was to investigate the effect of secretory factors of human mesenchymal stem cells on gentamicin-induced acute kidney injury in rats.Material and Methods: Firstly, an experiment was designed to achieve a standard animal model. Animals were randomly divided into four groups. One group was considered as control which received no injection. Three other groups were treated with intraperitoneal 100 mg/kg gentamicin for six consecutive days. Then one of the three groups was injucted with conditioned media of human mesenchymal stem cells and other group was injucted with medium without secretory factors intraperitoneally for three consecutive days. Finally, in the third, fifth and eighth days after gentamicin injection, kidney and blood samples from all animals were collected for histological and biochemical analysis.Result: Results, except in the special case, showed no significant decrease in the amount of blood biomarkers of animals in the group receiving the condition media of human mesenchymal stem cells compared to two another groups. The improvement of tissue damage, however, has been observed in the number of days.Conclusions: These findings suggest that secretory factors of human mesenchymal stem cells have aprotective effect against gentamicin-induced nephrotoxicity.

Abstract: 

Aim: Present study was done to assess the effects of Av gel on some molecular aspects in wound healing process of mouse damaged skin.Material and Methods: 36 Male mice balb/c were used. Animals were divided into 3 groups; negative control (without wound), sham-operated (wound treated with physiological serum) and experimental (wound treated with Av gel extract). In each group, two equal full-thickness wounds were made on the mice back sides. Experimental group received daily a dose of Av gel extract on the top of wound sites (without bandage) for a period of 16 days. On 8th and 16th post wounding day, for the purpose of EGF receptor gene expression evaluation and lipid peroxidation (LPO), skin and blood serum sampling were done respectively and data were analyzed by Repeated measures ANOVA at significant level p<0.05Results: Av gel extract treatments increased EGF receptor gene expression in the wound area. Wound healing percentage was also increased in experimental groups compared to sham group. It was also found that the amount of LPO end-product in blood sera was reduced to Av treated mice in the end of test.Conclusion: Av gel extract can induce EGF receptor gene expression in mice damaged skin, and play a pivotal role in wound healing process.

Abstract: 

Aim: Hepatopancreas as a vital organ whose function changes during life, effects clearly on histological structure. Since there is no study about this matter this study has been done.Material and Methods: In this study fifty healthy mature vannamei with the average length of 12±1 cm and weight of 18 ± 0.5 gr were caught from breeding farms. After adaptation, shrimps were kept in two aquavarium with same condition while one group was kept hungry for 5 days and another group (control group) was fed during the same time. Then 20 hepatopancreas from each group were taken and held in Davidson solution fixative. The routine procedures of preparation of tissues were followed and the cutted paraffin blocks were stained, and studied under light microscope.Results: Microscopic findings, revealed kinds of M, E, B and R cells. The number of cells, size of lobules and the weight of hepatopancreas in non-hungry and hungry samples were measured and examined with statistical analysis. Results showed reduce in number and area of hepatopancreas tubules especially in resorptive cells than to other cells.Conclusion: According to the findings, weight and area of hepatopancreas, size of cells and tubules in two groups was significance with statistical analysis.

Abstract: 

Aim: The somites are epithelial blocks of mesodermal cells differentiating to sclerotome and dermomyotome and notochord or axial mesoderm plays a major role in somitic cell differentiation to sclerotome. This study was aimed to evaluate the role of the notochord in sclerotomal differentiation of the somites in vitro.Material and Methods: In this experimental study, isolated notochords from chick embryo were encapsulated in alginate beads and co-cultured with somites for six days. In control group, the somites were cultured alone. Newly isolated and non-cultured somites were considered as somites on day 0. Eventually, morphology of cultured somitic cells was evaluated by inverted microscope and then RT-PCR method was used to analyze the sclerotomal and dermomyotomal gene expression profile.Results: In comparison with control group, the somitic cells co-cultured with notochord had highly differentiation potential and showed mesenchymal morphology with numerous slender processes. Gene expression profile of co-cultured somitic cells indicated upregulation of Pax1 and BMP4 and downregulation of MyoD, presenting sclerotomal cell characteristics.Conclusion: Embryonic notochord can induce in vitro sclerotomal differentiation in somites through upregulation of genes involving in this differentiation.

Abstract: 

Aim: In this research, the antibacterial effects of essential oil of two medicinal plants, Allium sativum and Rosmarinus officinalis were studied on Staphylococcus agalactiae, S. aureu and Escherichia coliMaterial and Methods: Essential oils were prepared from department of medical plants of Arak University. The antibacterial properties of the essences were investigated by disk diffusion method in different concentrations of the essences (10, 30 and 50%). MIC and MBC were determined by using different concentrations of essences. Antibacterial activity of Penicillin, Bacitracin and Erythromycin was determined with the discs that were impregnated with antibiotics.Results: All concentrations of the essential oil (10, 30 and 50%) had antibacterial effect. Antibacterial effects decreased according to reduction of essential oil concentrations. There were no significant difference between Allium sativum and Rosmarinus officinalis in MIC and MBC. There were, however, significant differences among mean zones of inhibition found in essential oils and antibiotics (Penicillin, Bacitracin and Streptomycin) at 10% concentration.Conclusion: According to antibacterial effect of Allium sativum and Rosmarinus officinalis essential oils on major bacterial mastitis pathogens in cow these plants has the potential to be evaluated as an alternative or adjunct to antibiotics to treat bovine mastitis.

Abstract: 

Angiogenesis is a necessary process in many physiological and pathological events such as tumor growth. Therefore angiogenesis therapy can be a strategy to cancer treatment. It is important to identify different aspects of tumor’s angiogenesis. Formation of functional vessels within tumors is essential for its growth and progression. The induction of new blood vessel growth by tumors is mediated through different molecules. A balance between pro-angiogenic and anti-angiogenic growth factors strictly control the process. This fact has led to design of therapeutic agents against tumor angiogenesis. Recently, endogenous (such as interleukin -8) and exogenous angiogenesis inhibitors (such as: Avastin) have been described. Furthermore because of the low mutagenic potential of endothelial cells, tumor’s vessel do not show resistance to the effects of many of these compounds. In addition, it is recognized that anti-angiogenesis and anti-cancer tumors can be more effective by combination of factors than the use of either alone. This article would review different aspects of angiogenesis in tumors. Since tumor growth is dependent on the development of blood vessels in the tumor, inhibition of angiogenesis maybe considered an appropriate treatment for cancer.
