Abstract:

Background: Two Dimensional Electrophoresis (2DE) is the most commonly and useful separation technique in proteomics. Each proteome snapshot becomes a protein profile. By means of this technique, several proteins are studied simultaneously. Methods: In this study, by use of (2DE) method, the differences of two profiles of Buthidae endemic scorpions, A.Crassicauda known as "black scorpion" and "O.doriae" yellow scorpion, were investigated. Results: For A.Crassicauda scorpion there were about 20 spots (peptides) in 6.2-8.2 pH ranges and molecular weight was less than 3 to 14 kDa and O.doriae scorpion 30 peptides, in 6.3-8.5 pH ranges, 1 to 45 kDa that fractionated and identified. Conclusion: By this method, the field of bioinformative data bank from Iranian endemic scorpions' venom could be prepared. By making change of any effective factors on scorpion venom, considerable results due to influence of the factor on determining kind of venom can be achieved and studied.

Background: Coagulase Negative staphylococci (CoNS) are nosocomial pathogens. The main objective of the study was to compare the performance of disk diffusion, E-test and agar screening methods with Real-time PCR technique for detection of methicillin resistance in coagulase negative staphylococci (MRCoNS), using Taqmanâ Real-time PCR for mecA DV WKH3 JROG VWDQ GDUG' FRP SDULVRQ DVVD. Methods: 88 coagulase negative Staphylococcus isolates were identified out of 284 Staphylococcus isolates collected from Hajar and Kashani hospitals-shahrekod, Iran. Methicillin resistance strains were identified by several methods: Disk diffusion, Agar screening, E-test and Real-time PCR. The results of the tested methods ZHUH FRP were compared with those of the Real-time PCR by Chi square or Fisher's exact tests. Results: Of the 88 coagulase negative Staphylococcus isolates tested, 46 isolates (52.3%) were mecApositive and 42 isolates (47.7%) were mecA-negative. The results of all the tested methods had a statistically significant agreement with those of Real-time PCR. The E-test was 100% sensitive and specific for mecA presence. The sensitivity and specificity of oxacillin agar screen (6.0 mg/ml) method were 96% and 98%, respectively and the sensitivity and specificity of oxacillin Disk diffusion method were 91% and 90%, respectively. Conclusion: In the present study, E-test is proposed as the best phenotypic method. In case economic issue matters, the oxacillin agar screening method (6.0 mg/ml), which is suitable for the detection of MRCoNS due to its accuracy and low cost, is recommended.

Abstract:

Background: Obesity-related disorders characterized by insulin resistance such as metabolic syndrome, hypertension, coronary artery disease and type 2 diabetes are associated with decreased adiponectin levels. The purpose of the present study was to determine association of lipid profile and obesity markers with serum adiponectin in type 2 diabetes adult males. Methods: For this purpose, forty one adult males with type 2 diabetes were enrolled in this study randomly. Blood samples were collected after a 10-12 hour overnight fasting for measuring serum glucose and lipid profile and adiponectin. The anthropometrical indices were measured too. A multivariate linear regression analysis used to predict relation between variables. Results: The data analysis showed that circulatory adiponectin had significant negative association with glucose, triglyceride, total cholesterol, visceral fat and cholesterol/HDL ratio (P£0.05). There were no significant correlation between adiponectin level and HDL and LDL.Conclusion: Our findings support negative relationship between serum adiponectin and cardiovascular risk factors in diabetic patients.

Abstract:

Background: Inactivity is a leading contributor to chronic health problems. Physical activity (PA) is an important element in maintaining the health and functional ability in the population and has favorable effects on lipid profile in adults. Here, we examined the effects of pedometer-based PA (step/day) in healthy middle age men. Methods: Height, weight, body mass index (BMI), and daily oxygen consumption (VO2peak) were measured in 27 active and sedentary healthy middle aged men (40-65 yr). Subjects wore a pedometer throughout the day for three consecutive weeks, and average steps per day (physical activity volume) were measured. Fasting serum concentrations of total cholesterol (TC), triglyceride (TG), high density lipoproteincholesterol (HDL-C), and low density lipoprotein-cholesterol (LDL-C) were measured. Results: Mean of Daily steps in the Active group (AG) (12632±3957 steps/day) was significantly higher than sedentary group (SG) (5347±1631 step/day) (p<0.001). Also There was a significant difference in daily oxygen consumption in the 2 groups, with higher values found in the AG (p<0.001). TG and TC concentrations in the AG were significantly lower than SG (p<0.05). TC/HDL-C and LDL-C/HDL-C ratios in the AG were significantly lower than SG (p<0.05), whereas, HDL-C and LDL-C concentrations were not different in the two groups (p>0.05). A significant correlation was found between average steps per day and TG (-0.424, P=0.027), TC (-0.389, P=0.045), TC/HDL-C (-0.469, P=0.014), HDL-C/LDL-C (-0.390, P=0.044), and Daily Oxygen Consumption (-0.853, P=0.000).Conclusion: It seems that, healthy middle-aged men, who have more ambulatory activity (average of 12, 500 step/day), have significantly better blood lipids' profile.

Abstract:

Aging is associated with reduced bone mass in men and women. Women loose about 35% of their cortical bone and 50% of trabecular bone since middle of 2nd decade of life. Subsequently, bone mass reduction increases fracture in women. Femoral bone fracture is associated with chronic low back pain, scoliosis, limited range of movement and more hospital admission. Prevalence of osteoporosis is increasing. It has been estimated that direct therapeutic costs and indirect costs due to reduced economical productivity of osteoporotic patients will increase 2 or 3 times. Moreover, it is also estimated that more than half of femoral bone fractures' of the world will occur in Asia. Therefore, there is need to further explore different aspects of osteoporosis locally. This book reviews physiology, diagnostic methods and different treatments of osteoporosis. It also discusses pathophysiological and diagnostic complexities of osteoporosis. Osteoporosis essentials are presented in combination with its new therapeutic findings. Therefore, the book could be used as a comprehensive tool for improving and updating current knowledge of readers about osteoporosis.
