Abstract:

The aim of this study was to investigate the effect of chitosan as a natural preservative. Farmed fish were washed and filleted. In this study, 3 types of chitosan were investigated: acid-soluble chitosan, water-soluble chitosan and oligosaccharide chitosan. The fillets were immersed in the above solutions for 30 seconds, and then removed, and after 2 minutes, the immersion was repeated. Control samples were uncovered. The fillets were then placed at room temperature (20 ° C) for 2 hours to form a coating on the fillets. 192 pieces of 100 g fish fillets were divided into 4 treatments and each treatment was divided into 4 times: treatment one as a control (without preservative), the second treatment containing 1% acid-soluble chitosan, the third treatment containing 1% watersoluble chitosan and the fourth treatment containing 1% Chitosan oligosaccharides. All samples were stored in refrigerator at 4± 1 ° C and their chemical parameters (Peroxide, pH, Thiobarbituric acid, Total Volatile Nitrogen) were measured on (0, 4, 8 and 12) days. All the indices of treatments had increasing trend over time. Only in the pH index of 1% acid-soluble chitosan had an irregular trend. 1% acid-soluble chitosan had the most positive effect in all treatments. Due to increase in chemical indices in all treatments, the addition different of chitosans to fish fillets could prevent or reduce them.

Abstract:

The present study intends to assess the effects of Na diformate oral use with citric acid on some skin mucosal immunity indices (total protein, total immunoglobulin (TIg), lysozyme, protease, esterase, and alkaline phosphatase (ALP)) of rainbow trout following experimental challenge with Yersiniosis agent (Yersinia ruckeri). Subsequent to the determination of the 7-day median lethal dose (LD50) of Y. ruckeri, fish were stocked in eight groups and two repetitions. The acidifiers separately with a dose of 0. 2%, and combined 0. 1+0. 1% was added to the commercial diet. Five days after inoculation, feeding of fish with acidifiers commenced, and following 15 days, the mucus samples were gathered from the skin of anesthetized fish to measure immune indices. Results showed that the 7-day LD50 of Y. ruckeri was 8. 91×105 CFU/ml. In the main experiment, findings indicated the significant effect of Na diformate on the increased levels of mucoid total protein and proteolytic activity of healthy or diseased fish. The acidifiers-contained diet because of potentially having synergistic effect caused inducing the mucus TIg. It seems that the orally use of Na difirmate in 0. 2% of the diet separately or combined with citric acid, at least for 15 days, can improve the cutaneous mucus immunological indices that its result is an increase in the survival rate of farmed rainbow trout.

Abstract:

The aim of this study was to compare the accumulation of heavy metals copper, zinc and cadmium and to investigate the changes of metallothionein as a biomarker of contamination in the gill tissue of the ghost crab Ocypode saratan in the two time periods before Monsoon (May) and after Monsoon (November) in mongrove forests. Chabahar and Gwater Bay was done in 2019. The results of the study showed that in both pre-Monsoon and post-Monsoon seasons the pattern of metal accumulation in the gill tissue of the ghost crab is zinc> copper> cadmium. The mean concentrations of copper, zinc and cadmium were obtained in the pre-monsoon and post-monsoon seasons at Chabahar station, 173. 6 ± 3. 83, 186. 43 ± 3. 1, 0. 095 ± 0. 008, respectively and 156. 7± 4. 15، 175. 13 ± 2. 8, 0. 026 ± 0. 003 and in Gwater station the average concentrations of copper, zinc, cadmium in the two pre-Monsoon and post-Monsoon seasons were respectively 237. 03 ± 6. 4, 231. 8 ± 1. 8, 0. 048 ± 0. 003 and 205. 9 ± 6. 34, 253. 2 ± 2. 13, 0. 037 ± 0. 015 μ g/g dry weight. The mean levels of metallothionein in pre-monsoon and post-monsoon season in Chabahar station were 2. 02 ± 0. 01 and 0. 1 ± 0. 06, respectively, and in goiter station were 2 ± 0. 02 and 0. 12 ± 0. 02, respectively. Comparison of metallothionein levels between Chabahar and Gwater stations was not significant (P<0. 05). But in both stations separately between the two pre-Monsoon and post-Monsoon seasons, there was a significant difference (P<0. 05). Correlation between metallothionein and metals showed that there was no correlation between metallothionein and the studied metals. Therefore, according to the observations, changes in the concentration of metallothionein in this species are not affected by heavy metals and can be due to changes in the environmental conditions of the seasons.

Abstract:

In the present study, the structural and antioxidant properties of fucoidan extracted from macroalgae (Sargassum and Padina) from the Persian Gulf coast were evaluated. The fucoidan was extracted by ethanol and hot water and after calculating the extraction yield, DPPH radical scavenging activity, ferric reducing antioxidant power (FRAP), scanning electron microscopy (SEM), and its monosaccharides by HPLC method were examined. The yield of fucoidan in Sargassum (4. 2 ± 0. 00%) was higher than Padina (2. 98 ± 0. 28%) (P < 0. 05). The IC50 of DPPH free radical scavenging in fucoidan extracted from Sargassum and Padina were 0. 1 and 0. 14 mg/ ml, respectively, which were higher than BHT as a commercial antioxidant (P < 0. 05). The rate of FRAP in both samples increased with an increase in the concentration of fucoidan. The SEM results showed that fucoidan from both macroalgae had strong structural cohesion and irregular surfaces, but Padina had more surface protrusions. The glucose, mannose and xylose sugars in different amounts were detected by the HPLC method, in which the amount of glucose and xylose sugars were higher in the fucoidan derived-Sargassum (P < 0. 05). According to the results of the present study, the fucoidan extracted from the Sargassum and Padina can be a good alternative to an industrial antioxidant in food.

Abstract:

In this study, the morphological variation of Oxynoemacheilus bergianus was studied in the different rivers of the Namak Lake and Caspian Sea basins using traditional morphometric method. For this purpose, a total of 76 specimens were collected from eight river systems and after fixation into 4% buffered formalin, transfered to the laboratory, a total of 31 morphological characteristics were measured using digital calipers. After standardization, the morphometric data were analyzed using multivariate analysis including principal component analysis (PCA), canonical variate analysis with p-value obtained from MANOVA (MANOVA/CVA) and cluster analysis (CA). The results showed significant differences in 24 traits between the studied populations (P<0. 05), which anal fin depth and the ventral-anal fin distances were main discriminative ones. CVA analysis was able to separate the studied populations. Also, CA placed the Gharesu and Sefid populations in a clade and separate from other populations. The observed differences may be related to phenotype plasticity in response to environmental conditions.

Abstract:

The main focus for this study was to compare and evaluate the quality changes and to determine the shelf life of frozen Scomberomorus commerson fillet using quick and slow freezing methods. Scomberomorus commerson fillets were frozen by slow and quick freezing methods and kept in the freezer at-18° C for 180 days. Then proximate analysis, chemical test (Total volatile base nitrogen; TVN, Thiobarbituric acid; TBA), drip loss, fatty acids profile and sensory analysis were assessed. Generally, Ash content, TBA, and TVN, drip loss, SFA values were significantly increased during storage time whereas the moisture, protein, fat, PUFA and MUFA were significantly decreased (P<0. 05). The ratio of TVN, TBA and drip loss was found to be higher in slowly as against quickly frozen fish. The PUFA (19. 71), MUFA (23. 40) and ω 3 (14. 69) of the quick samples was higher than that of the slow samples (16. 99, 22. 64, 12. 34%, respectively). Regarding sensory analysis, both samples were in acceptable condition but the quality of quick samples was better than slow frozen samples. The quick freezing method was generally much more influent on quality parameters than slow freezing method.

Abstract:

Primary production is a key indicator in the evalution of aquatic ecosystems that can directly affect carbon sequestration. Due to climate change and environmental dynamics, this component has changed spatially and temporally in different ecosystems. Therefore, to understand and monitor these changes, the most important influential parameters include; sea surface temperature, chlorophyll-a and algal bloom on this component and their changes were analyzed based on the time series. Images related to Chl-a, SST and NPP from January 2003 to December 2018 were used by MODIS sensor and VGPM model to estimate NPP in the study area. Aftar pre-processing and extracting the time series algorithm, the trend of variation was determined using the mann-kendall and theil-sen procedure. Then, using enhancing false color composite, algal blooms in the Persian Gulf and the sea of Oman were identified and the amount of chl-a concentration in the algal bloom area and outside the area was determined. The results of trend analysis with theil-sen procedure showed that the rate of change for chl-a is decreasing in all regions except the fifth region. The highest amount was observed in the fifth region (-0. 19), which corresponded to the rate of NPP. The results also show that the highest amount of NPP in the first region in March and the lowest in June and July (540 and 690 gC/m2 /day), which corresponds to the amount of chl-a concentration. Despite the apparent correlation, there is no cause-and-effect relationship between SST and chl-a as well as chl-a and NPP. In this way, factors other than chl-a can be considered for algal bloom and NPP.
