Abstract: 

Background: Skeletal muscle as an endocrine tissue is involved in the regulation of metabolic activity, production and secretion of hormones including myokines. The aim of the present study was to investigate the effect of eight weeks of aerobic training combined with ethanol consumption on plasma lipid profile and glucose levels, triglyceride content and mayonectin, irisin and leptin gene expression in the gastrocnemius muscle in male rats. Methods: A number of 32 rats with a weighing average of 200 ± 10 g were divided into four groups control, aerobic training, ethanol with dose of 4 g/kgbw, and ethanol +aerobic training. At the end of the period, values of lipid profile and plasma glucose, the amount of triglyceride of the gastrocnemius muscle and the relative levels of mayonectin, irisin and leptin gene expression were evaluated. Data were analyzed using two-way ANOVA. Results: The plasma triglyceride levels in the aerobic training and ethanol+aerobic training groups were significantly decreased compared to the control group (P≤ 0. 05). The gastrocnemius muscle triglyceride values were significantly increased in the ethanol +aerobic training (P≤ 0. 0001) and aerobic training groups (P≤ 0. 01) compared to the control group. The results showed that aerobic training significantly increased myonectin gene expression in aerobic training group (P≤ 0. 05), but the expression of irisin and leptin genes did not change significantly in different groups. Conclusion: Aerobic training during the eight-week was able to improve lipid content, especially plasma triglyceride and skeletal muscle triglyceride, and possibly regulate body metabolism by altering the levels of myokines, especially myonectin.

Abstract: 

Background: The purpose of this study was to compare the effect of eight weeks of resistance training with and without the use of aqueous extracts of barberry and saffron on serum levels of RBP4, glucose, insulin and insulin resistance in obese mice. Methods: In this study, 80 mice with a mean weight of 41± 2 g were exposed to high fat diet for 10 weeks. Then, they were randomly assigned to 8 resistance training groups (10 n), resistance and saffron (10 n), saffron (10= n ), Resistance and barberry (n =10), barberry (n =10), saffron and barberry (10n =), resistance training with extract of saffron and barberry (10n =) and control group (n = 10). The resistance training program included raising the mice from the ladder with weight (three sets and 5 repetitions each) three times a week for 8 weeks. The consumption of saffron and barberry extracts was done at 0. 08 for total weight of mice for 8 weeks. After 14 hours of fasting, the initial blood sample was taken at 5 ml of the left ventricle of the rats and RBP4, glucose, insulin and insulin resistance levels were measured. One-way analysis of variance was used to compare the data between groups. All calculations were considered at P≤ 0. 05 Results: The results showed that there was a significant difference in the levels of RBP4 (P = 0. 002), glucose (P = 0. 03), insulin (P = 0. 02), and insulin resistance (P= 0. 004) There are different research groups. Follow-up test results showed that RBP4 levels decreased in the resistance training group compared to the barberry and control groups. In saffron group, resistance to barberry and control group decreased. The saffron group also decreased compared to the saffron and barberry training groups. Conclusion: The findings of this study emphasize the effect of resistance training, the use of saffron extract and barberry extract on reducing the metabolic effects of obesity by reducing RBP4 and reducing glucose, insulin and insulin resistance.

Abstract: 

Background: Obesity and type 2 diabetes can impair the function of important cellular pathways. Activation of the mTOR pathway results in regulation of the SREBP1 protein for metabolism and regulation of adipose tissue. The aim of this study was to investigate the effect of 4 weeks of high intensity interval training on the content of mTOR and SREBP1 in adipose tissue of type 2 diabetic rats. Methods: In this experimental study, 12 to 2-month-old male Sprague-Dawley rats weighing 300 20± 20 g were selected and after being diabetic by induction of STZ and nicotine amide, randomly divided in two groups, diabetic training (6 rats) and diabetic control (6 rats). Exercise group training 4 days a week for 4 weeks according to the training HIIT; The control group had no exercise program. Independent t-test and dependent t-test were used for data analysis. Results: There was no significant change in mTOR protein content (p=0. 12); But the SREBP1 protein content (p=0. 001) increased significantly. The weight of control group (P=0. 0001) and HIIT group (P=0. 010) showed a significant increase. Blood sugar in the control group also increased significantly (P=0. 0001), but HIIT mice did not show a significant change (P = 0. 14). Conclusion: 4 weeks of HIIT training did not significantly change weight, blood glucose and mTOR protein content. But it did increase the SREBP1 content, so factors such as duration and intensity of training should be adjusted in order to achieve the best results when administering HIIT.

Abstract: 

Background: Although a positive association between type 2 diabetes and breast cancer has been reported, an association with gestational diabetes mellitus (GDM) is less clear. This study aimed to assess the relationship between the history of GDM and the incidence of BC a long term population based study. Methods: To conduct this research, 4076 women aged 20-50 years who participated in the first phase of Tehran-Lipid-and-Glucose-Study, who had at-least one pregnancy were included. Each participants were asked about the history of GDM every 3 years. They was evaluated about the occurrence of breast cancer (BC). Cox-survival analysis was used to measure the Hazard-ratio (HR) for both groups with and without history of GDM Results: The study population included 886 women with prior GDM and 3, 909 women without history of GDM. The median duration of follow-up among women with and without the history of GDM were 12. 12 and 11. 62 years, respectively. The incidence of BC was 0. 64 per 1, 000 year-olds with confidence=interval of 95% (0. 44-0. 91). Among them, 4 cases were reported in women with GDM and 26 cases in women without GDM. (Incidence of 0. 36 per 1000 person-years with confidence=interval 95% (0. 11-0. 99) in women with prior GDM and 0. 73 per 1, 000 person-years with confidence=interval 95% (0. 49-1. 1) in women without a history of GDM. Conclusion: Our study showed that the history of GDM is not an independent risk factor for breast cancer.

Abstract: 

Background: Type II diabetes is a chronic inflammatory condition that is associated with a combination of genetic and environmental factors. Tumor necrosis factor alpha or TNF-α as an adipocyte cytokine, which affects the signaling pathway of insulin, can contribute to insulin resistance in type 2 diabetes patients. Considering the importance of epigenetic changes in multifactorial diseases, this study aimed to investigate TNF-α promoter methylation in patients with type 2 diabetes. Methods: This study was performed on 61 patients with type 2 diabetes and 31 non-diabetic patients. The Groups were matched in terms of demographic characteristics. The lipid profiles were measured by standard kits. TNF-α promoter methylation levels were measured by bisulphite treatment method, Nested PCR and sequencing. Results: There was no association between TNF-α promoter methylation gene promoter and type 2 diabetes in the studied groups. Also, there was no association between TNF-α promoter methylation in diabetic and non-diabetic groups between males and females. Conclusion: The epigenetic changes in cytokines that contribute to insulin resistance in type 2 diabetic patients seem to be ineffective in peripheral blood samples, and other tissues may need to be investigated in this regard.

Abstract: 

Background: Peripheral neuropathy is one of the most common problems in diabetic patients. The increased risk of Diabetic Foot Ulceration (DFU) and amputation would be a complication of diabetic neuropathy. The aim of this study was to compare the DFU healing in different severity classification of neuropathy. Methods: This is a retrospective study that was conducted over a two-year period from April 2016 to March 2018 according to the information of patients records with Diabetes Mellitus (DM) referred to clinic of diabetes and metabolic disorders of Endocrinology and Metabolism Research Institute of Tehran University of Medical Sciences. Wound healing criteria including area, depth and healing duration were studied. Accordingly, changes in the area and depth of wounds were evaluated and reported during the first, third and sixth months after baseline. Data were analyzed using descriptive and inferential statistics using SPSS software version 16. Results: The results of the study of patients with neuropathic ulcer showed that males and age group of 56 to 65 years had the highest frequencies. In addition, most of these patients suffered from type 2 DM (79%). The rate of wound healing, which was measured by area and depth of wound in three time periods, differed in different severity classification of neuropathy; at mild level of neuropathy the area and depth of wound decreased faster but in severe neuropathy, duration of wound healing in both mentioned criteria has increased. Regarding to the increase in the duration of DM, the healing time increased too. However the rate of wound healing decreased with increased age (p-value = 0. 001). Conclusion: Evidence suggests that early identification of neuropathy can reduce the incidence of DFU and amputation. Due to the slow healing of the size and depth of the wound in diabetic patients with severe neuropathy, it is necessary to prevent DFU by conducting preventive care and educational interventions.
