Abstract: 

Background and Objective: The anti inflammatory, analgesic, antioxidant of Sambucus ebulus L. have been reported in several studies. This study was done to assess the repelling property of the methalonic and hexanic extracts of Sambucus ebulus L. against the Culex pipiens.Methods: In this experimental study, Sambucus ebulus L. collected from the natural inhabitants of Mazandran province in northern Iran. Methalonic and hexanic extraction were provided from the leaf and fruit of Sambucus ebulus L. Concentration of 50 mg/kg, 100 mg/kg and 250 mg/kg was prepared.0.4 ml of the extract prepared and was spreed on the albino skin area of 4×6 cm2. After 30 minutes the number of the mosquito (Culex pipiens) bites on the skin was recorded. N, Ndiethyl-3 methyl benzamide was considered as positive control.Results: The highest repelling property of the Sambucus ebulus L. belonged to the concentration of 250 mg/kg of leaf and fruit extraction. The highest repelling effect was 80% and 66.8% for leaf methalonic and hexanic extract, respectively. The highest repelling effect was 84% and 72% for fruit metalonic and hexanic extract, respectively (P<0.05).Conclusion: The methalonic extract of Sambucus ebulus L. had higher repelling efficiency compared to the hexanic extract. The fruit extract also had better effect than the leaf extract.

Abstract: 

Background and Objective: Ferula assa foetida (Ferula assa-foetida L.) is an herbaceous wild plant native to Iran which is used in the traditional medicine for treating stomach and intestinal disorders. This study was done to determine the antidiarrheal effect of hydroalcoholic extract of Ferula assa foetida in rat.Methods: In this experimental study, Wistar adult male rats randomly allocated into negative control, positive control and interventional groups. Animals in negative control groups were recived normal salin orally. Animals in positive control groups were recived Atropine (0.1 mg/kg/bw) for evaluation of intestinal propulsive movement and Loperamide (3 mg/kg/bw) for evaluation of diarrhea. In interventional group 1, 2 and 3 animals were received hydroalcoholic extract of Ferula assa foetida 100, 200 and 400 mg/kg/bw, resepectively. One hour after administration of extracts and medicine diarrhea induced using castor oil in animals. Induced diarrhea, intestinal propulsive movement and intestinal fluid accumulation were evaluated in rats.Results: Gavage of the extract (5 g/kg) did not produce any toxic effect in rats. The mean peristaltic index in Gavage for doses of 100, 200 and 400 mg/kg/bw, control and atropine groups was 97.00%, 65.88%, 62.23%, 86.19% and 52.86%, respectively. The extract at the lowest dose in combination with atropine was significantly reduced peristaltic index rather than of the atropin alone (P<0.05). The extract produced a non-significant reduction in the volume of intestinal fluid accumulation and propulsive movement in the castor oil-induced intestinal transit in rats. In a dose-dependent manner, the extract delayed the onset of diarrhea. Loperamide and highest dose of extract (400 mg/kg/bw) produced a significant reduction in the frequency of defecation and severity of diarrhea (P<0.05).Conclusion: The hydroalcoholic extract of F. assa foetida showed anti-diarrheal activity due to its inhibitory effect on intestinal fluid accumulation.

Abstract: 

Background and Objective: Iron oxide nanoparticles have wide applications such as MRI contrast agent and drug delivery. Nevertheless, their effects on human health have not been fully investigated yet. After cellulose, chitin is one of the most abundant organic materials in nature which is widely used in food industry, cosmetics, agriculture, medicine and the environment. This study was done to evaluate the effect of iron oxide nanoparticles coated with chitosan on renal functional indeces in rat.Methods: In this experimental study, 60 adult female Wistar rats were allocated into 10 equal groups. Concentrations of 50, 100 and 150 mg/kg/bw from chitosan, iron oxide nanoparticles and chitosan coated nanoparticles were intraperitoneally injected into 9 groups and animals in control group were received normal saline. Blood samples were collected directly from the rat heart in the days 15 and 30 post after injection and renal functional indeces including urea, creatinine, uric acid, sodium, potassium and total protein were measured.Results: There were no significant differences in the level of urea, creatinine, uric acid, sodium, potassium and total protein in the groups whom received chitosan-coated iron oxide nanoparticles compared to control. There was no mortality during the study time.Conclusion: Short-term using of iron oxide nanoparticles coated with chitosan does not create any toxicity in the rat kidney.

Abstract: 

Background and Objective: Iron oxide nanoparticles, including nanoparticles is important in industry and medicine. Nanoparticles affect on detoxification of environmental pollutants such as Pesticides and chlorinated organic solvents. This study was done to evaluate the short term effect of Fe2NiO4 composite nanoparticle on kidney function indeces in wistar rats.Methods: In this experimental study, Twenty four Wistar rat were randomly allocated into three groups, including: control, treated groups 1 and 2. Animales in control, treated groups 1 and 2 were received 0.5cc of saline, 0.5cc of solution containing 100, 200 ppm Fe2NiO4 for 7 days, respectively. Uric acid, ceratinine and urea (BUN) were measured at day 2, 7 and 14.Results: BUN level in treated groups 1 and 2 significantly reduced in comparison with control group at day 7, 14 after intervention (P<0.05). Uric acid level in treated groups 1 and 2 significantly increased at day 7 and 14.2 week after intervention, the mean creatinine levels in treated group 2 group significantly reduced in compare to the in treated group 1 and controls (P<0.05).Conclusion: It seems that the application of Fe2NiO4 nanoparticles in biological system has no toxic effect on the kidney function indeces.

Abstract: 

Background and Objective: Liver diseases accompanied with growing of obesity in children. This study was done to evaluate the effect of aerobic exercise and vitamin C intake on liver transaminases activities in 8-11 years obese girls.Methods: In this quasi-experimental study, 28 obese girls were randomly divided into four equal groups including control, exercise, supplement (500 mg vitamin C tablet, dailly) and combined (500 mg vitamin C, daily plus exercise) groups. Aerobic exercise was consisted of exercise movements at 50% to 70% maximal heart rate and 3 sessions per week for 8 weeks. Fasting blood samples were collected before and 48 hour after the last intervention. Serum transaminases activities were measured by enzymatic colorimetric method.Results: 8 weeks of aerobic exercise, vitamin C and the combined intervention were associated with a significant reduction in Alanine aminotransferase (ALT) and Aspartate aminotransferase (AST) activities (P<0.05) whereas there was no effect on the Alkaline phosphatase activity. All these interventions were associated with significantly greater reduction in the ALT and AST activities ratio in comparision with control group. Combined intervention induced more reduction on percent of variables changes compared with other interventions (P<0.05).Conclusion: It seems selected aerobic exercise and vitamin C intake may induce their protective effect in obese girls via improvement in liver function.

Abstract: 

Background and Objective: Gender differences in biobehavioral responses to environmental stressors and experience of psychological stress should be identified. This study was done to evaluate the changes of the level of salivary cortisol under psychological stress and its relationship with rumination and personality traits.Methods: In this case-control study, for 45 medical students, The NEO Personality Inventory-Short Form and emotional control questionnaire (ECQ) were filed two months before the final examination. Saliva samples were taken from students in the non-stress (control) and examination stress conditions. Salivary cortisol levels were measured by ELISA method.Results: Gender differences were not observed in the level of salivary cortisol under psychological stress. Significant difference was observed between the mean of salivary cortisol in the non-stress and under examination stress conditions. Positive correlation was found between traits of neuroticism (P<0.05) and rumination (P<0.05) with salivary cortisol as well as negative correlation between the traits openness to experience (P<0.05) and angery control (P<0.05) with salivary cortisol. Neuroticism, rumination and angery control may predict a substantial variance (32%) of salivary cortisol under exam stress.Conclusion: Psychological stress leads to increase in the secretion of salivary cortisol unrelated to gender. Subjects with different personality traits are prone to cortisol responses to stress based on their particular character.

Abstract: 

Background and Objective: Micro RNAs (or miRNAs) control gene regulation and different biological processes in various tissues and therefore play an important role in various diseases. In some cases, either a single nucleotide polymorphisms (SNPs) in miRNAs or in complementary sequences in their target mRNAs play significant role in human diseases. In this study, the relationship between rs531564 G>C in mir-124-1 with the susceptibility to type 2 diabetes in the Iranian population was examined.Methods: In this case-control study, 173 individuals affected with type 2 diabetes and 162 healthy individuals were selected. Extracted DNA from peripheral blood was amplified by a pair of relevant primers and then digested by BsmAI restriction enzymes. The obtained electrophoretic patterns were used for genotyping.Results: The genotype frequencies of GG, GC and CC for rs531564 in the patient group were 0.92, 0.06 and 0.02 respectively, 0.96, 0.04 and 0.00 in the controls. Statistical analysis showed no significant difference between the two groups regarding the genotype frequencies, however the allelic frequencies were significantly different between those groups (P<0.05).Conclusion: There was no genotype difference between diabetes and healthy individuals, but the allelic C is related with type 2 diabetes among Iranian population.

Abstract: 

Background and Objective: Dietary intake of b-carotene and b-cryptoxanthin may be associated with reduce the risk of insulin resistance, due to their antioxidant properties. The aim of this study was to determine the relation between dietary intake of b-carotene and b-cryptoxanthin and the risk of insulin resistance in adults.Methods: In this prospective cohort study, 938 (421 men and 517 women), aged between 19 to 82 years were selected from among participants of the Tehran Lipid and Glucose Study in Iran. Dietary intake of β-carotene and b-cryptoxanthin were determined using a valid and reliable food frequency questionnaire. Fasting serum glucose and insulin were measured at base line and again after a 3-year of follow up. Logistic regression models were used to estimate the occurrence of insulin resistance across tetiles of b-carotene and b-cryptoxanthin with adjustment for potential confounding variables.Results: The mean age of participants was 40.7±12.1 years. b-carotene was inversely associated with insulin resistance after adjustment for confounder variables (95% CI=0.25–0.72, OR=0.42, P-value for trend=0.01). Also, an inverse association was found between dietary intake of b-cryptoxanthin and insulin resistance risk (95% CI=0.30-0.84, OR= 0.51, P-value for trend=0.01).Conclusion: There was inverse association between dietary intake of b-carotene and b-cryptoxanthin and risk of insulin resistance in adults.

Abstract: 

Background and Objective: Although lifestyle-related factors have separately been examined in relation to functional gastrointestinal disorders (FGIDs), there is no epidemiologic data on the combined association of lifestyle factors with these conditions. We aimed to examine how combinations of several lifestyle factors were associated with functional dyspepsia (FD), its symptoms and gastro-esophageal reflux disease (GERD) in a large group of Iranian adults.Methods: This descriptive -analytic study was conducted on 3363 Iranian adults (19-70 yr), whom were working in 50 health centers across Isfahan province in Iran during 2012. We used easy non-random sampling to select participants. The “healthy lifestyle score” for each participant was calculated by summing up the binary score given for five lifestyle factors, including dietary habits, dietary intakes, psychological distress, smoking and physical activity. A dish-based 106-item semi-quantitative validated food frequency questionnaire, General Practice Physical Activity Questionnaire, General Health Questionnaire and other pre-tested questionnaires were used to assess the components of healthy lifestyle score. A validated Persian version of ROME III questionnaire was used, to assess functional gastrointestinal disorders.Results: The prevalence of FD and GERD among study participants was 14.5 and 23.6%, respectively. After adjustment for potential confounders, we found that individuals with the highest score of healthy lifestyle had 79 and 74% lower odds of FD (95% CI: 0.05-0.92, OR: 0.21, P=0.03) and GERD (95% CI: 0.09-0.69, OR: 0.26, P=0.01), respectively, compared with those with the lowest score. They were also less likely to have early satiation (95% CI: 0.11-0.73, OR: 0.28, P=0.001), postprandial fullness (95% CI: 0.09-0.50, OR: 0.22, P<0.001) and epigastric pain (95% CI: 0.21-0.92, OR: 0.44, P=0.03). In addition to the combined healthy lifestyle score, low levels of psychological distress, a healthy diet, healthy dietary habits and non-smoking were separately associated with FGIDs (P<0.05).Conclusion: This study showed that adherence to a healthy lifestyle was associated with lower odds of GERD, FD and its symptoms in this group of Iranian adults. Individual lifestyle-related factors were also associated with these conditions.

Abstract: 

Background and Objective: Helicobacter pylori are one of the most common gastrointestinal bacterium infections. This study was done to compare the urease test with Giemsa staining to detection of Helicobacter pylori infection in patients with gastric ulcer and gastritis.Methods: This descriptive – analytic study was conducted on 601 patients' diagnosed with gastric ulcer and gastritis, whom under went endoscopy in 5th Azar hospital, Gorgan, Iran during 2008-12. Rapid urease test was immediately taken during endoscopy. Pathological examination, Giemsa staining on paraffin blocks and haematoxylin and eosin stain were done to assess either presence or absence of Helicobacter pylori.Results: Based on biopsy results, Gastritis and gastric ulcer were diagnosed in 80.69% and 19.3%, respectively. In gastritis and gastric ulcer patients, there was a significant differences between urease test (91.5% and 90%, respectively) in comparison with Giemsa staining (91.5% and 90%, respectively) (P<0.05).Conclusion: The study revealed that in primary diagnosis and screening of role of Helicobacter pylori in patients with gastric ulcer and gastritis, urease test is suitable than giemsa staining.

Abstract: 

Background and Objective: A high percentage of patients with multiple traumas sustained at least from an orthopedic problem. One of the high frequent lesions was femoral shaft fracture. The aim of this study was to determine the amount of bleeding and the need for blood transfusion in femoral shaft fractures and risk factors.Methods: This descriptive, retrospective study was done on 84 patients with femoral shaft fractures whom were operated in Imam Khomeini Hospital in, Sari, in northern Iran during 2012-15. Age, sex, comorbidities, type of fracture, hospitalization period, pre and post-operative hemoglobin and blood unit's loss were recorded for each patient.Results: In 43 patients (51.19%) plate and in 41 patients (48.80%) intramedullary naling were used for treatment of fractures.13 (15.47%), 33 (39.28%) and 37 (44.04%) of patients were received one, two and three blood units, respectively. There was no significant relationship between energy intensity and age. There was significant relationship between the intensity and the type of fracture (P<0.05). A significant correlation existed between blood unit transfusion with sex, fracture type, hemoglobin before surgery and fracture energy intensity (P<0.05).Conclusion: Based on the results of this study, it is recommended for hip fracture at least 3 units of blood should be reserved.

Abstract: 

Background and Objective: Calcitonin is a small peptide hormone including 32 amino acids and 3.4 KD molecular weight which is produced by the parafollicular cells of the thyroid gland in respond to increasing calcium ions in serum. This peptide is used for adjuvant therapy of osteoporosis, Paget' s disease and hypercalcemic shock. In this study, the heterologuse expression of calcitonin was done in Escherichia coli.Methods: In this experimental study, the thioredoxin fusion partner was added to n-terminal of the Salmon calciton in order to increase its stability by the synthetic biology. The recombinant construct was transformed and over expressed into Escherichia coli BL21 (DE3) host cell.Results: SDS-PAGE analysis showed the over expression of recombinant protein after IPTG induction.Conclusion: In this study, the construct including fused Salmon calcitonin gene with thioredoxin was cloned. The SDS-PAGE result showed the stable expression of fused calcitonin.

Abstract: 

Background and Objective: Tumor size results in hypoxic and acidic environment leading to the production of several types of growth factors required for the formation of blood vessels. Afterwards, metastasis of cancerous cells occurs via blood vessels. Therefore angiogenesis inhibition can be a new way of cancer treatment. This study was done to determine the bioinformatics analysis to predict potential Micro-RNAs inhibiting processes of angiogenesis in cancer.Methods: In this descriptive study, micro-RNAs that are able to connect to MMP genes involved in tumor angiogenesis (MMP1-2-3-8-9-10-11-13) were detected using miRwalk database. Effective Micro-RNAs selection was based on the number of binding sites in 3' UTR genes. MicroRNA data base was used to find sample base pairing sequences.Results: mir-1302, mir-516a, mir-512, mir-511, mir-516b and mir-548 were determined with the most number of binding sites in genes involved in angiogenesis.Conclusion: MicroRNAs are worthy options for cell culture and laboratory examination in order to find new ways to prevent the development of cancer by angiogenesis inhibition.

Abstract: 

Background and Objective: Hypericin is found in different species of Hypericum genus, as a main compound with antimicrobial, antiviral, nonspecific kinas inhibition and dopamin b-hydroxilase inhibitoring effects. This study was done to compare the hypericin content, antioxidant, antimicrobial and cytotoxic activities of Hypericum perforatum L. from three geographic regions of Iran.Methods: In this descriptive study, Hypericin content of aerial parts of H. perforatum L. was assessed using UV-Vis spectrometric method. Antioxidant activity was measured using DPPH and b-carotene bleaching assay. Cytotoxicity was evaluated via brine shrimp lethality assay. Antimicrobial activity was determined using inhibition zone diameter evaluation via disc diffusion method and measuring minimum inhibitory concentration (MIC) value.Results: Hypericin content of aerial parts of H. perforatum L. from Qom, Golestan and Kurdestan provinces were 673, 1223 and 1568 ppm, respectively. Antioxidant and cytotoxic activities in samples from Kurdestan was more than samples from Qom and Golestan. Antimicrobial activity, as far as the number of sensitive microorganisms was evaluated. In this way the order of Golestan>Kurdestan>Qom was exhibited, however the extract of the plant from Kurdestan had the highest activity for two staphylococcus species with the inhibition zone diameter of 17 and 19 mm for S. aureus and S.epidermidis, respectively and MIC value of 250 mg/mL.Conclusion: Hypericin content was more from samples of Kurdistan province with cold climate. Antimicrobial, antioxidant and cytotoxic activities of aerial parts of all samples were high. There is a relationship between hypericin content of aerial parts of H. perforatum L. and biological activities.

Abstract: 

Background and Objective: The blood–brain barrier (BBB) is considered to be the main barrier to drug transport into the central nervous system. In this study, the capability of biopartitioning micellar chromatography (BMC) using the mixed micellar system of Brij-35/sodium dodecyl sulfate (Brij-35/SDS, 85: 15 mol/mol) has been studied to predict pharmacokinetic parameter (BBB penetration ability) of 14 basic drugs.Methods: In this descriptive-analytical study, the potential of BMC using mixed micellar system (Brij-35/SDS, 85: 15 mol/mol) in 0.04 M at physiological pH 7.4 was evaluated to predict pharmacokinetic parameter (BBB penetration ability) of 14 basic drugs. The regression model for the prediction of blood-brain distribution coefficient is derived from the multiple linear regression analysis using the training set in mixed micellar mobile phase. Also, the predictive ability of model was evaluated for a prediction set of 5 compounds (Chlorpromazine, Mianserin, Propranolol, Cimetidine, and Thioridazine). The fair R2 indicates good stability and predictive ability of the developed model for the drugs not included in modeling.Results: The relationship between the BMC retention data of 14 basic drugs and their log BB parameter showed a good statistically model (R2=0.822, F=25.42, SE=0.225, R2CV=0.781).Conclusion: This study points out the usefulness of mixed micellar solution of Brij-35/SDS, 85: 15 (mol/mol) in BMC as a high-throughput primary screening tool that can provide key information about the blood-brain distribution of basic drugs in a simple and economical way.

Abstract: 

Background and Objective: Varicella zoster virus (VZV) can cause a moderate disease in children, but with high risk of serious disease or death in children with acute lymphoblastic leukemia (ALL) Vaccination from infection can be safe, immunogenic, and effective in children with leukemia. This study was done to evaluate the IgG and IgM antibodies against VZV in children with acute lymphoblastic leukemia.Methods: The descriptive-analytic study was performed on 66 children who were on chemotherapy in Dr Shigh hospital in Mashhad, Iran during 2012. Patients were receivied VZV vaccine. The title negative or positive serum sample for IgG-anti-VZV avidity was determined by using a test kit before and after injection of vaccines VZV for considering the efficacy of vaccines on pediatric patients.Results: Title serum sample IgG were positive in 32.6% patients, these children have history of chicken pox disease and to be safe against of VZV.78.3% of the patients were negative for IgG antibody and sensitive against of VZV.Conclusion: Children with ALL receiving chemotherapy are sensitive to chicken pox disease according to negative IgG titer against VZV.
