ECHIUM AMOENUM STIMULATE OF LYMPHOCYTE PROLIFERATION AND INHIBIT OF HUMORAL ANTIBODY SYNTHESIS

Abstract

 Background: In Iranian traditional medicine, Echium amoenum, is used as a native MEDICINAL plant. Objective: To investigate the effect of Echium amoenum extract on humoral and cellular immune response. Methods: Hydroalcoholic extract of Echium amoenum was prepared and the proliferative responsiveness of peripheral blood and thymic lymphocytes to mitogen and alloantigen was determined by LYMPHOCYTE PROLIFERATION assay and two-way mixed lymphocyte reaction, respectively.Humoral ANTIBODY PRODUCTION to sheep red blood cell (SRBC) in mice was investigated by multiple intraperitonealy administration of the extract (1-100 ?g/ml) 24 and 48 hours before and after immunization of a group of mice with SRBC. The primary and secondary antibody titers to SRBC were determined by hemagglutination assay.Results: A strong increase in allogenic response was obtained at 10 ?g/ml of the extract (stimulation index of 1.81, p< 0.01). Echium amoenum showed no direct stimulatory activity on either lymphocytes or thymocytes in proliferation assay. However, the extract at concentrations of 50-400 ?g/ml showed a co-stimulatory effect on mitogenic LYMPHOCYTE PROLIFERATION. The highest stimulation index was obtained at concentration of 200 ?g/ml (1.44, p< 0.02). Despite these immunostimulatory effects, the extract showed a significant dose related decrease in the humoral antibody response (p<0.05). The mean antibody titer log2 for doses of 25, 50 and 100 mg/kg of the extract in the primary response ranged from 5.1?0.7 to 6.5?0.5 compared to the negative control (8.5?0.9). The corresponding data for secondary response was 4.6?0.7 to 6.6?0.5 compared to 10.8?0.7 in negative control.Conclusion: Although, the extract showed the capacity to augment LYMPHOCYTE PROLIFERATION in the presence of mitogen or alloantigen, humoral antibody synthesis in both primary and secondary response was inhibited.
