THE EFFICACY OF MORPHINE ON MURINE (BALB/C) CUTANEOUS LEISHMANIASIS

Abstract

 Background and objective: CUTANEOUS LEISHMANIASIS (CL) is a potentially disfiguring parasitic disease which is endemic in many parts of the world. The evidence for optimal treatment of CL is uncongenial. The development of a compound used for the topical treatment of CL is an attractive option for overcoming the problems with toxic and expensive currently used antileishmanial drugs. Evidence is certainly about a variety of immune responses in the healing process of CL. The immune modulatory effect of MORPHINE also has been known in clinical medicine for decades.There are some evidences that opioids could be members of the cytokine family and the regulation of immune and inflammatory responses is dependent on the functions of cytokines. The objective of this study was to determine the effects of MORPHINE as an immunomodulator on the healing process of CUTANEOUS LEISHMANIASIS in BALB/C mice.Materials and methods: Forty BALB/C mice were randomly divided into five groups. All mice were subcutaneously inoculated each with a number of 2.5x106 promastigotes ofLEISHMANIA MAJORon the base of their tails. Groups A and B received topical MORPHINE, since the first and fifth weeks of inoculation, respectively, for three weeks daily. In the same way, groups C and D received weekly intraperitoneal MORPHINE for three weeks; the control group (E) received no any treatment.Then the diameters of cutaneous lesions were measured weekly using a vernier-caliper.Results: The measurement of the cutaneous lesions showed increasing size in all mice receiving topical or intraperitoneal MORPHINE. The mean diameters of cutaneous lesions were significantly greater in the fifth week comparing to the first week. There was no significant difference in diameters of cutaneous lesions between treatment groups (A, B, C, D) and control group (E) at the same time.Conclusion: Topical application and intraperitoneal injection of MORPHINE were not effective in decreasing the size of CL in BALB/C mice. Other studies with different doses of MORPHINE could be helpful to show possible recovery. Different kind of mice such as Souris or Hamsters might reveal different results.
