Abstract

Aquired immunodeficiency syndrome (AIDS) is caused by HIV11,2 viruses. Three major structural proteins gp41, gag(p24) , gp36 are the most common used for EIA diagnostic techniqce for detection of specific antibodies in sera and plasma in infected individuals. In this Study, recombinant proteins gp41, p24, gp36 were expressed in E.coli. Rec gp41 and Rec gp36 contain N-terminal part of HIV1portion of the transmembrane protein gp41 and gp36 from HIV1 and HIV1,2 respectively as well as common core gag (P24) protein. These proteins were highly purified more than 98.5% and used as a solid phase antigen for immunoreactivity evalution. The accuracy and specifity of anti HIVI,2 antibody detection were evaluated with representative panel of positive and negative and seroconversion sera. HIV1 positive collection included: HIV1,2 seroconversion panel (BBl) # PRB 934 (n=3). Iranian collection HIV1 positive (309). Russian Refrence Standard Panel HIV1 positive (n=15) and weak positive HIV1 (n=4) and HIV1,2 positive collection from Oganda (n=3). HIV1/2 negative Iranian collection, (n=500), 30 negative samples from high risk people, 10 samples from autoimmune patients, 10 samples from multifarious woman and 10 samples from Hepatitis C patients. Russian national Reference HIV11/2 negative panel (n=15). Sensitivity and accuracy are the most important for a screening test. This greatly depends on antigen quality. Sensitivity was found 100% and specifity, 99.8% and reactivity for seroconversion panel was seventy days of bleeding.
