QUANTITATIVE ANALYSIS OF HIPPOCAMPAL NEURON NUMBER AND SIZE FOLLOWING PRENATAL ADMINISTRATION OF BOSWELLLA SERRATA GUM RESIN IN ADULT RATS

Abstract

 INTRODUCTION: We have previously shown that maternal administration of BOSWELLIA SERRATA gum resin during gestation can increase LEARNING and MEMORY of adult male rats. The hippocampal formation is known to be involved in the control of this ability. The purpose of the present study was to determine whether prenatal consumption of BOSWELLIA SERRATA had delayed effects on the numbers and individual soma volume (size) of granular and pyramidal cell layers in the adult rat HIPPOCAMPUS.METHODS: Two-month-old male Wistar rats whose mothers were given oral aqueous extract of the BOSWELLIA SERRATA (0.1 g/kg/day) during gestation (3 weeks) were anesthetized and transcardially perfused with phosphate buffered solution of 4% formaldehyde and 1% glutaraldehyde (n=8). The Optical Fractionator technique was used to determine the total number of hippocampal neurons and individual somal volume measurement was performed by the use of the Rotator method.RESULTS: Prenatal administration of BOSWELLIA SERRATA gum resin was found to have no effect on the total number of hippocampal neurons. Two-tailed t-test revealed only significant increase in the volume of hippocampal neurons in CA3 (P<0.01).DISCUSSIONS: tereological data of this study provide a neuroanatomical basis that may be relevant to the reported enhancement of LEARNING and MEMORY abilities in offspring. The constituents and precise mechanisms responsible for the efficacy of BOSWELLIA SERRATA are still a matter of debate and remain to be clarified.
