Abstract

Background: The increasing use of MEDICAL HERBs and their products and the fact of mutagenicity and oncogenicity of some of their components has justified further research about these effects of herbal products. This research was done for evaluation of effects of PROSTATAN and SANKOL as two herbal products on HUMAN CHROMOSOMES in Shiraz in 2000.Materials and Methods: Peripheral blood lymphocytes of healthy normal cases were exposed to different concentration levels of SANKOL and PROSTATAN in RPM1-1640 culture medium in this experimental study (Invitro). Frequency of polyploid cells, bicentromeric chromosome and chromatid breaks were observed. Hydroquinone as positive control and distilled water as negative control were used. The effect of ethanol as a solvent was also evaluated.Results: Abnormal metaphases severely increased using 0.2% and 0.5% concentration of SANKOL and PROSTATAN. Mean chromosome abnormalities following the use of PROSTATAN and SANKOL were 2.9% and 5.3%. Frequency of polyploid abnormalities and chromatid breaks was significantly higher by using SANKOL versus PROSTATAN. The frequency of chromosome abnormality following the use of both drugs was considerably more in comparison with that in the negative control group.Conclusion: As PROSTATAN and SANKOL cause increased chromosomal in vitro abnormalities, further in vivo studies on the effects of these drugs are recommended due to their frequent usage and prescription during pregnancy.
