PREDICTION AND COMPARISON OF PHARMACOLOGIC AND SUPRAPHARMACOLOGIC DOSES OF DIGOXIN ON NORMAL AND PITHED RAT HEARTS BY USING A COMPUTEIDZED SOFTWARE

Abstract

 BACKGROUND AND OBJECTIVES: In this study, capability of special COMPUTERIZED SOFTWARE for determining of DIGOXIN effects in pharmacologic and suprapharmacologic doses on cardiac hemodynamic functions were investigated.METHODS: Different doses of DIGOXIN (0.1-100mg/kg) were injected (IV) in normal and PITHED RATs by using the software and the effects on cardiac homodynamic functions including arterial blood pressure (ABP), heart rate (HR) and heart force (HF) were evaluated and compared.FINDINGS: In normal rats, DIGOXIN had no significant effects on ABP and HR while using 0.5-100mg/kg of the agent produced a marked and significant increase in HF (p<0.001) where the effect was dose-dependent at the doses of 0.5-20mg/kg (r=0.98). Similarly, in pithed test rats, DIGOXIN (0.5-100mg/kg) showed significant elevation in HF versus pithed control group (p<0.001) without any statistically significant changes in ABP and HR. Higher doses of DIGOXIN (50-100mg/kg) caused severe and lethal ventricular arrhythmia in both normal and PITHED RATs. Compared to the PITHED RATs, ABP and HR were significantly higher than the normal rats (p<0.001).CONCLUSION: The software based on pharmacologic properties of DIGOXIN may be able to predict cardiac effects of suprapharmacologic doses of the agent in normal and PITHED RATs.
