EVALUATION OF ANTIMICROBIAL EFFECTS OF VARIOUS ORGANIC AND AQUEOUS EXTRACTS OF DATURA INNOXIA MILLER

Abstract

Datura innoxia has long been used by various nations for many therapeutic purposes such as analgesic. In this study, antimicrobial activity of various extracts of the plant aerial parts was investigated.Aerial parts of D. innoxia were collected in Shahran highlands, northwest of Tehran city, in June 2008 and shade dried. Powdered plant specimen was then extracted by the maceration method using aqueous and organic solvents. The dried extracts were reconstituted using DMSO at both 10 and 100 mg/ml and antimicrobial activities were evaluated against an array of 17 Gram-positive (GPB) and Gram-negative (GNB) bacteria as well as 2 fungi (Candida albicans and Aspergillus niger) by the cup-plate method. Minimum inhibitory concentrations (MIC) of the extracts were also determined by the Agar dilution technique against bacteria.Based on our finding, the antibacterial activity of extracts tested was in the following order: methanol > water > chloroform > petroleum ether > ethyl acetate. Methanolic and aqueous extracts producing clear zones with diameters >20 mm showed to have strong activity on some GPBs including both sensitive and resistant staphylococci, Micrococcus luteus and Bacillus subtilis. Other extract had no significant antibacterial activity. On the other hand, MIC values obtained were >200 μg/ml against all bacteria. Furthermore, these extracts showed no activity against test fungi.In spite of antibacterial activity observed for methanolic and aqueous extracts against GPBs in the screening stage, MIC values indicate that these extracts have no considerable antimicrobial activity. This finding disagrees with some previous reports on remarkable activity of this plant on GPBs.
