INVESTIGATION OF CONTROLLED RELEASE OF INDOMETHACIN DRUG FROM 2-HYDROXYPROPYL METHACRYLATE POLYMERIC CARRIERS

Abstract

 IN THE LAST DECADES MUCH ATTENTION HAS BEEN DIRECTED TO SPECIALITY POLYMERS FOR BIOMEDICAL APPLICATION. ONE PARTICULAR APPROACH TOWARDS THE IMPROVED USE OF DRUGS IS THE DESIGN OF POLYMER-DRUG CONJUGATES OR POLYMERIC PRODRUGS. THE CHEMICAL ATTACHMENT OF LOW MOLECULAR WEIGHT DRUGS TO SYNTHETIC OR NATURAL POLYMERS HAS BEEN FREQUENTLY INVESTIGATED AS A MEANS OF IMPROVING THE EFFICACY OF DRUG CONTROL RELEASE DEVICES THROUGH A CONSTANT BUT PROLONGED RELEASE OF DRUGS WITH MINIMUM SIDE EFFECTS [1]. INDOMETHACIN IS A NONSTEROIDAL ANTI-INFLAMMATORY ANALGESIC USED IN THE TREATMENT OF DISORDERS SUCH AS RHEUMATOID ARTHRITIS, ANKYLOSING SPONDYLITIS, AND OSTEOARTHRITIS [2].THE AIM OF THE PRESENT PAPER WAS TO SYNTHESIZE, CHARACTERIZE AND IN-VITRO HYDROLYSIS OF POLYMERIC SYSTEMS CONTAINING HYDROLYSABLE ESTER BONDS LINKED TO INDOMETHACIN FOR CONTROLLED DRUG RELEASE. FIRST, INDOMETHACIN LINKED TO 2-HYDROXYPROPYL METHACRYLATE IN THE PRESENCE OF N, N-DICYCLOHEXYLCARBODIIMIDE BY ESTER METHODOLOGY. THE RESULTED MATERIAL WAS THEN COPOLYMERIZED WITH 2-HYDROXYETHYL METHACRYLATE AND METHYL METHACRYLATE BY UTILIZING AZOISOBUTYRONITRILE AS AN INITIATOR TO GIVE DRUG-POLYMER CONJUGATES. THE OBTAINED MATERIALS WERE CHARACTERIZED BY FT-IR, NMR, AND ELEMENTAL ANALYSIS TECHNIQUES TO CONFIRM THEIR STRUCTURES. THE HYDROLYSIS OF DRUG-POLYMER CONJUGATES WAS CARRIED OUT IN CELLOPHANE MEMBRANE DIALYSIS BAGS CONTAINING AQUEOUS BUFFER SOLUTIONS (PH 1, 7.4 AND 10) AT 37OC FOR 24 H. DETECTION OF HYDROLYSIS SOLUTION BY UV SPECTROSCOPY AT SELECTED INTERVALS SHOWED THAT NAPROXEN CAN BE RELEASED BY HYDROLYSIS OF THE ESTER BOND BETWEEN THE DRUG AND POLYMER BACKBONE. THE RELEASE PROFILES INDICATED THAT THE HYDROLYTIC BEHAVIOR OF POLYMERIC PRODRUGS IS STRONGLY BASED ON THE POLYMER HYDROPHILICITY AND THE PH VALUE OF THE HYDROLYSIS SOLUTION.
