Abstract:

The aim of the present study was to investigate the potential of nanoemulsion formulation for topical delivery of Clobetasol propionate (CP) using algal oil (containing omega-3 fatty acids) as the oil phase. CP has anti-inflammatory, immunomodulatory and antiproliferative activities. However, its clinical use is restricted to some extent due to its poor permeability across the skin. Algal oil was used as the oil phase and was also exploited for its anti-infl ammatory effect along with CP in the treatment of infl ammation associated with dermatitis. Nanoemulsion formulations were prepared by aqueous phase titration method, using algal oil, tween 20, PEG 200 and water as the oil phase, surfactant, co-surfactant and aqueous phase respectively. Furthermore, different formulations were subjected to evaluate for ex-vivo permeation and in-vivo anti-inflammatory, irritation and contact dermatitis studies. The optimized nanoemulsion was converted into hydrogel-thickened nanoemulsion system (HTN) using carbopol 971 and had a viscosity of 97. 57 ± 0. 04 PaS. The optimized formulation had small average diameter (120 nm) with zeta potential of-37. 01 mV which indicated good long-term stability. In-vivo anti-infl ammatory activity indicated 84. 55% and 41. 04% inhibition of inflammation for drug loaded and placebo formulations respectively. The assessment of skin permeation was done by DSC and histopathology studies which indicated changes in the structure of epidermal membrane of skin. Contact dermatitis reveals that the higher NTPDase activity in the treatment with the CP-loaded nanoemulsion could be related to the higher anti-inflammatory effect in comparison with placebo nanoemulsion gel.

Abstract:

Fatty acid profile and proximate composition of horse mackerel (Trachurus mediterraneus), sole (Solea solea) and black scorpion fish (Scorpaena porcus) caught from the Marmara Sea were determined both in the summer (June-July-August) and winter (December-January-February) seasons. Fish is the best source of omega-3 (ω-3) and contains plenty of omega-6 (ω-6) fatty acids. ω-3 fatty acids provide so many health benefits and help to protect against diseases. In this study, the lipid content of horse mackerel was higher than the other investigated fish species. Total ω-3 and eicosapentaenoic acid (EPA) + docosahexaenoic acid (DHA) of horse mackerel were the highest in summer (1705 mg 100g-1) despite having higher lipid content in winter, which could be explained by the nourishment that fish takes in different seasons. Considering the recommended intake, EPA+DHA of horse mackerel was more than enough to meet the daily requirement. The ω-3 content of black scorpion fish and sole were relatively low in comparison with horse mackerel in both seasons. The ratio of ω-6: ω-3 fatty acids of all species was also very low (max. 0. 358) which provides advantage in daily nutrition. Docosapentaenoic acid (DPA) and α-linolenic acid (ALA) were also found higher in horse mackerel in winter. Instead of taking supplements, fish itself should be consumed to benefit all long chain ω-3 polyunsaturated fatty acids (PUFA) and other nutrients. For a healthy diet, consuming horse mackerel routinely seems to fulfil EPA and DHA needs, besides having high quality protein.

Abstract:

Background and Objectives: Eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) have preventative and therapeutic effects against various disorders, especially cardiovascular and inflammatory conditions. Considering the advantages of using nanoliposomes as carrier systems of bioactive compounds, this study was aimed to the development nanoliposomes containing vitamin E loaded with DHA and EPA and evaluation of their physical stability.Materials and Methods: Preformed nano-liposomes were prepared based on dipalmitoyl-phosphatidylcholine (DPPC) with vitamin E using a modification thin film method and sonication. Loading of EPA and DHA into nano-liposomes was done by incubation. To improve stability, vitamin E was added to some formulae. After formulation, a comparative study was performed between them through evaluation by Zetasizer, transmission electron microscopy (TEM), differential scanning calorimetry (DSC), and calculating the percent of DHA and EPA entrapment efficiency. Physical stability study was carried out by investigating the change in the vesicles’ encapsulation efficiency, particle size, size distribution and shape when stored at 4, 30 and 40oC for 3 months. Results: The results showed that vitamin E-based nanoliposomal formulations successfully entrapped EPA and DHA with high encapsulation efficiencies (81.96±1.4% and 89.1±0.6%), respectively, and appropriate particle size (82±0.8 nm) and PDI (0.13±0.02) were achieved for the optimized formulation. The results of the physical stability tests of the optimized nanoliposome carriers of DHA and EPA incorporated with vitamin E were suitable after a 3-month storage at 4oC. DSC and TEM results confirmed vitamin E–DPPC: Omega 3 interactions and vesicular dispersion phenomenon of DHA and EPA.Conclusion: The findings of this study showed that incorporation of vitamin E in nanoliposomal formulation had significant protective and stabilizing effects on the DHA and EPA loaded in bilayer membrane lipid and its physical stability. The combination of preservatives and vitamin E in liposome containing EPA and DHA was not more effective on the physical stability of liposome formulation than the control formulation.
