Abstract: 

Background: One of the most common problems in diabetic patients is muscle atrophy. Therefore, the present study aimed to investigate the synergistic effect of endurance training with probiotic supplementation on Atrogin-1 and MuRF-1 genes gene expression in the soleus muscle of diabetic rats. Methods: In this study, 32 male Wistar rats were randomly divided into one normal control (NC) and four diabetic groups: diabetic control (DC), diabetic supplement (SDC), diabetic training (TD), and diabetic supplement training (STD). The training protocol was performed with 60 to 65% of maximum speed reached five days a week for four weeks. At the same time, rats took two grams of probiotic dissolved in 30 ml of water daily. Expression of Atrogin-1 and MuRF-1 genes was measured by the qReal-Time PCR method. Data were analyzed by two-way analysis of variance at the significant level of P≤, 0. 05. Results: Atrogin-1 gene expression was significantly reduced in TD (P=0. 001) and STD (P=0. 000) groups compared to DC group. There was a significant difference between TD and STD groups in the expression of the Atrogin-1 gene (P=0. 028). MuRF-1 gene expression was significantly reduced in TD (P=0. 04) and STD (P=0. 01) groups compared to DC. But there was no significant difference between TD and STD groups in MuRF-1 gene expression (P=0. 36). Conclusion: It seems that performing the aerobic exercise with probiotic supplementation is more effective in reducing the expression of the Atrogin-1 gene than any of these interventions alone. However, aerobic exercise with probiotic supplementation does not have a synergistic effect on reducing MuRF-1 gene expression in the soleus muscle of diabetic rats compared to aerobic exercise alone.

Abstract: 

Background: Empagliflozin, selective glucose-sodium inhibitor of the latest drugs in the treatment of type 2 diabetes. Diabetes induced hypogonadism disrupts sexual function. There is a direct relationship between reducing blood glucose and reduced libido. In this project, the anti-diabetic drug Empagliflozin in addition to treatment has been studied, in terms of effect on sexual function. Methods: Type 2 diabetes with injection of 60 mg/kg streptozotocin per body weight was create intraperitoneally. The primary diabetes was treated with two doses of 10 and 25 mg/kg Empagliflozin per body weight. Sexual hormones, biochemical parameters and lipid profile were measured. Evaluation of Sperm parameters and morphological studies were performed on testicular tissue, pancreas and epididymis. Data were analyzed by one-way ANOVA in SPSS software version 22 and P˂, 0. 05 was considered as a significant level. Results: Empagliflozin significantly increases the quality, survival and natural state of sperm head and tail. Empagliflozin reduces weight in diabetic rats. Empagliflozin Without increasing hyperglycemia by reducing blood glucose affects steroidogenesis, maintains fertility strength in testicles, and increases the level of sex hormones. Empagliflozin significantly increases insulin secretion. Empagliflozin maintains and integrity of pancreatic beta cells. Empagliflozin has no effect on lipid profile. Conclusion: Empagliflozin in addition positive effects on the treatment of diabetes in the early stage improves sexual function in adult rats.

Abstract: 

Background: Dysfunction of Schwann cells by diabetes has far-reaching consequences for the structure and function of peripheral nerves. The aim of the present study was to evaluate the effect of aerobic exercise on the Nrg1 / ErbB2 signaling pathway in male rats with diabetic neuropathic pain. Methods: Twenty-four 8-week-old male Wistar rats (weight range 204 ±,11. 3 g) were randomly divided into three groups (n = 8) including: Diabetic neuropathy (50 mg / kg streptozotocin injection), Diabetic neuropathy Exercise (30 minutes exercise aerobics intensity 15 m/min, 5 days per week for 6 weeks), and healthy controls. After confirmation of diabetic neuropathic pain by behavioral tests, aerobic exercise protocol was performed. The expression of Nrg1 / ErbB2 genes was measured by real-time technique. One-way analysis of variance and Tukey's post hoc test were used for statistical analysis. Results: Aerobic exercise reduced the nervous system's sensitivity to thermal hyperalgesia and mechanical allodynia. Induction of diabetes significantly reduced Nrg1 and ErbB2 gene expression. There was also a significant increase in the expression of the above genes in the exercise group compared to the diabetic neuropathy group (P<0. 05). Conclusion: It seems that aerobic exercise is effective in increasing the nerve repair process by activating growth signaling pathways in Schwann cells. It is recommended that aerobic exercise be used for diabetics to reduce neuropathic pain.

Abstract: 

Background: Diabetes is one of the most common chronic diseases in today's society. The aim of this study was to investigate the effect of endurance training and nettle consumption on the protein and gene expression of AKT, GLUT4 and insulin in male of muscle soleus of diabetic rats. Methods: After adaptation to the environment, 40 male Wistar rats were divided into 5 groups: control (C), diabetes (D), nettle+diabetes (UD), exercise+diabetes (ED) and exercise+nettle+diabetes (EUD). After developing and confirming the diabetes model by injection of STZ, rats in the exercise groups performed an endurance training protocol for eight weeks and rats in the nettle groups consumed nettle extract 5 days a week. After completion of the protocol and extraction of soleus muscle, RT-PCR method was used to measure gene expression, Western blot and immunohistochemically method was used to measure protein expression of glut-4 and AKT respectively and ELIZA method was used to measure glucose and insulin. Results: The results showed that the protein and genes expression of GLUT4 and AKT in the diabetic group compared to the control group was significantly reduced (P<0. 001) while the expression of these two variables was significantly increased in the ED and EUD groups (P<0. 001) that this increase was greater in the EUD group. The effect of nettle alone on the expression of these two variables was not significant. Glucose, insulin and insulin resistance increased significantly in the diabetes group, while these decreased significantly in the ED and EUD groups (P<0. 001). Conclusion: The synergist of the combination of nettle and exercise significantly improves the improve of diabetes, while each alone has little effect on diabetes-related variables.

Abstract: 

Background: Hypoglycaemic effects of Galega officinalis and silver nanoparticles are established. In the present study, the effects of silver nanoparticles synthetized by Galega officinalis extract were investigated on gene expression of TNF-α, , IL-6 and serum levels of liver enzymes in diabetes type 2. Methods: In the present study 20 male Wistar rats in 4 group(n= 5 in each group) weighing 180-200 gr were used. Control or nikotinamid and stroptozotosin induced diabetic rats receieved intraperitoneal injection of saline or 2/5mg/Kg silver nanoparticles synthetized by Galega officinalis extract or chemichal method for 14 days respectively. One day after the last injections, serum samples and adipose tissue were collected. Mean serum concentration of glucose, urea, creatinine, alanine amino transferase(ALT), and aspartate amino transferase(AST) were determined by spectrophotometry. Mean relative gene expression of TNF-α,and IL-6 were determined by method of real time PCR. Results: Mean serum levels of ALT and AST enzymes, glucose, urea significantly decreased in diabetic rats receiving chemichal or green silver naoparticles compared to diabetic group. Mean relative gene expression of TNF-α,and IL-6 significantly decreased in diabetic rats receiving chemichal or green silver naoparticles compared to diabetic ones. Conclusion: Both chemical and green synthetized silver nanoparticles may prevent hepatocyes damages and they may improve insulin resistance in diabetes type 2 partly via decresing pro-inflammatory factors.

Abstract: 

Background: Diabetes is one of the major health problems in Iran and about 4. 6 million adults suffer from this disease. Poor diagnosis of this disease has caused half of this number to be unaware of their disease. In recent years, along with the use of computers in data analysis and storage, the volume and complexity of data has increased dramatically. Methods: In health organizations, data play an essential role in the value of the organization. Therefore, data mining has become one of the most widely used processes in the field of health and disease diagnosis. In this study, the information of 768 laboratory clients in Tehran was kept confidential and the opinions of experts were used to identify the variables affecting the incidence of diabetes. Results: The findings indicate the study of 5 algorithms on the presented data, which by implementing 5 data mining algorithms J48, Bayes, Beginning, Cohen and simple clustering to classify the data, the efficiency of these algorithms in terms of speed and accuracy in calculations was evaluated. Conclusion: The data set for classification is the database of a laboratory, which includes 768 samples with 9 characteristics. Finally, J48 algorithm is recommended for data mining of diabetes due to high speed, acceptable accuracy and lack of sensitivity to raw data.
