Abstract:

Animal studies show that ethanol exerts amnesic effects in different models of memory such as inhibitory avoidance. Probiotics are food supplements and pharmaceutical substances prepared from living microorganisms that with enhancement of microbial equilibrium of intestinal microbiota, insert useful effects on the host (human/animal. The aim of this study was examination of TD4, the native Iranianprobiotic isolated from Traditional dairy product on the rat passive avoidance. In this study use as 96 male wistar rats with 200-250 grams weight range. For induced amnesia used as ethanol and for one mount administrated with1×109 cfu/ml of with or without probiotic bacteria. The step-through passive avoidance used in the present study to examine long-term memory in male Wistar rats. Results indicate that post-training sub cortical (SC) administration of etanol (1 mg/kg) induce amnesia. This amnesia was reversed in probiotic receiving groups. Results of this study showed that the lactobacillus brevisisolated fromTarkhineh dough, enhance the on passive avoidance learning of male Wistar rats that resiveetanol. Our data demonstrate the positive effect of this probeotic on learning and memory.

16 newborn calves were randomly assigned to four experimental groups (4 calves for each group). T1:control group, T2:1 gr day-1 bacterial probiotic, T3:2 gr day-1 yeast probiotic and T4:1 gr day-1bacterial +2 gr day-1yeast probiotic was added in their daily milk Consumption until 70 days of age.Blood samples were obtained from jugular vein to determination of blood parameters such as Glucose (GLU), Total protein (TP), Triglyceride (TG), Cholesterol (Cho), Calcium (Ca), Phosphorus (P), and Beta- Hydroxyl Butyric Acid (bHBA), at three times (present at birth, 40 and 70 days. Amount of cholesterol in Treat 3 at the end of period was lower than other treatments and the amounts of bHBA and Ca in Treat 3, 4 at the middle of period were higher than other treatments (p<0.05). Feed intake and weight gain in Treat 3, 4 were significantly higher than Treat 1, 2 in the fourth and fifth weeks after born (p<0.05). Only 3 cases of diarrhea in Treat 1 and one case in Treat 2 were observed, but they werentdiarrhea in the other group. In conclusion the results of this study indicated that comparison of bacteria and yeast probiotics until 40 days have the greatest effect and after this time, it can reduce the impact of changing the intake of probiotics is associated with the development of therumen.

Abstract:

Stress and its role in amplifying symptoms of disease, is one of the most important problem of mankind in developed counties. By identification of the roles of microflora in brains function, it seems that probiotics could be used for control and ameliorating of brain responses such as stress. So in this study, the effect of Lactobacillus rhamnosus LX-108, in ACTH secretion level was investigated in male wistar rats under chronic restraint stress (CRS). In this research 24 male wistar rats weighing 180- 200 gr randomly divided in to 3 groups: Negative control, Positive control (received CRS), and trail group (orally gavaged with 1 ml of probiotic suspension and received CRS). Rat’s weight was measured every week and animals were anesthetized with ether after three weeks, and blood samples were obtained directly from their hearts. ACTH concentrations of sera were measured by ELISA method with special kit. Indeed microflora profile was identified in fecal samples. Our results showed that administration of LX-108 can inhibit the induction of ACTH under CRS condition. Besides this probiotic resulted in increasing of intestinal Lactic acid bacteria and decreasing the Enterobacteriaceae level which induced in CRS condition. Fortunately administering of this probiotic inhibit weight loss induced in CRS condition. After clinical trials, Lactobacillus rhamnosus LX-108 and its probiotic products can be used for controlling of stress and its distracutive effects.

Abstract:

Breast milk as the only source of nutrition in the first months after birth, an important factor in the development of microorganisms that are present in the intestines of infants. Given the importance of lactic acid bacteria, especially lactobacilli in human health, molecular recognition and classification of these bacteria could be an effective step in applying them in Functional products. The purpose of this research, detection and identification of Lactobacillus strains present in the breast milk. In this study, the strains in milk samples were isolated after enrichment in MRS broth, After microscopic examination, the basic characteristics of bacteria was determined by biochemical tests. Finally, to identify the molecular basis Identification of 16SrRNA sequences and genomic fingerprinting by REP-PCR was performed. Gram positive and catalase negative bacilli and cocci, were coded. Finally the PCR products were sequenced, and the sequences were analyzed with BLAST software. Sequencing results were analyzed using Ez Taxon server, the results showed that the strains isolated type of Pediococcus lolii, Lactobacillus rhamnosus and Lactobacillus fermentum. Breast milk can be considered as a source of lactic acid probiotic bacteria that may play a role in preventing of infant infectious disease have in the other hand the present study demonstrates the efficacy of REP-PCR genomic fingerprinting with marker (GTG) 5 in examining phylogenetic relationships and genetic variation in bacteria.

Abstract:

Probiotics are microorganisms, which reduce fatigue, improve immune function and strengthen the digestive system would be a positive impact on human performance. Effect of Bacillus coagulans probiotic supplementation on the physical and mental condition of the soldiers in this study were assessed. The 40 soldiers who were divided into 2 groups. To a group of probiotic fruit juice contains 2×109 colonies coagulans probiotic supplement and the base material probiotic control group (placebo). During the 8 week and every 15 days before and after 100 meters lactate levels were measured from blood samples were taken and the mental condition of people in probiotics by using a questionnaire Profile of Mood States (POMS) assessed. Bacillus coagulans probiotic supplementation results showed a significant reduction in lactate levels increase in physical activity compared with the control (P<0.05). Also Test subjects better mood compared to the control group. The results of this study, the use of probiotics in improving people's physical and mental condition.

Abstract:

The purpose of research is comparison methods of cadmium telluride quantum dot and magnetic silica core shell for detection of aflatoxin B1 based on fluorescence resonance energy transfer (FRET) from cadmium telluride quantum dot to Rho123. Cadmium telluride quantum dot was synthesized by reflux method aquatic phase, high temperature, alkalin pH and under nitrogen flow. The thioglycolic acid is used as a stabilizing and capping agent. After quantum dot synthesis, bougthen anti aflatoxin B1 antibody immobilized on the surface of quantum dot. Subsequently, magnetic silica core shell by using FeCl3 and FeSO4 synthesized under nitrogen flow and alkalin pH and to increase TEOS solution. After preparation aflatoxin B1-albumin bioconjugate is labeled with Rho 123. The specific immune-reaction between the anti-aflatoxin B1 antibody on the quantum dot and the labeled-aflatoxin B1 brings the Rho 123 fluorophore (the acceptor), and the quantum dot (the donor (in close spatial proximity and causes FRET to occur upon photo-excitation of the quantum dot. In the absence of unlabeled aflatoxin B1, the antigen-antibody complex is stable and strong emission of quantum dot observe. In the presence of free aflatoxin B1, it will compete with the labeled aflatoxin B1-albumin complex for binding to the antibody- quantum dot conjugate so that FRET increasingly suppressed. There is a linear relationship between the decreased fluorescence intensity of Rho 123 with increasing concentration of aflatoxin B1 in the range of 0.01-0.06 μmol.mL-1.By using the magnetic silica core shell as a signal intensifier brings sensitivity of the system increased from 2×10-11 to 2×10-12 μmol.mL-1.
