Abstract: 

Background and Objective: Breast cancer is the fifth common cause of death in the world and is considered as the leading cause of cancer related death among females. HER2 is a surface antigen which is overexpressed in 30% of breast cancer cases. The aim of this study was cloning of HER2 receptor gene in pCDNA3.1\Hygro+ expression vector, expression of this protein in LL2 eukaryotic cell line and production of a breast cancer mouse model.Materials and Methods: The construct expressing HER2 gene was synthesized by RNA extraction, HER2 gene amplification and its cloning into pCDNA3.1\Hygro+ vector. The accuracy of cloning was approved by sequencing. In the next step, the construct was transfected into LL2 mouse cancer cells. To establish a stable cell line, transfected LL2 cells were cultured in hygromycine enriched medium for 1 month. The resultant cells had a very low growth rate. Having repeated the process three times, 106 cells were eventually produced. Injection of these cells into mice led to no tumor formation. Moreover, LL2 cells transducted by HER2 containing lenti-viruse were used collaterally to form tumors. Meanwhile, 106 transducted LL2 cells were injected subcutaneously into C57BL/6 mice. A tumor nodule was tangible 10 to 12 days post injection.Results: The HER2 gene was successfully amplified and cloned into pcDNA3.1\Hygro+. Injection of transfected and transduced LL2 cells into mice demonstrated that only the LL2 cells transduced by the viral vector pLEX HER2 were able to express human HER2 gene stably.Conclusion: By cloning and expression of HER2 on the LL2 cells, we could produce breast cancer model in mice expressing human HER2 on cells surface.

Abstract: 

Background and Objective: Psychosocial stresses are one of the causes of most clinical problemes of general health of societies. It can produce and intensify many diseases such as cardiovascular disorders. Studeise have shown this type of stress could produce free radicales and lead to oxidative stress. As regular exercise is effective in decreasing mortality from cardiovasulare dieases, immobility and obesity are the main risk factores for occurring of these diseases.Materials and Methods: Fifty rats were randomly divided into 5 groups: Control group, Heart failure group, Psychosocial stress and Heart failure group, Exercise and Heart failure group, and Psychosocial stress, exercise and heart failure group. Psychosocial stress was induced by change of cage-mate of animals. The rats in the training intervention groups were trained to run on a treadmill, 5 days a week for 4 weeks. Subcutaneous injection of isoproterenol (130 mg/kg) was used to induce experimental heart failure.Severity of heart failure was evaluated by functional parameters such as FS, LVEDd, LVESd, LVPWT and EF by echocardiography. Plasma levels of malondialdehyde (MDA) and pro-oxidant/antioxidant balance (PAB), as oxidative parameters and corticostrone level were also evaluated in rat serum.Results: Psychosocial stress increased levels of PAB, MDA, corticostron and LVEDd and decreased EF, FS and LVPWT (P<0.05). Protective effects of regular exercise was shown with decreased levels of PAB, MDA, corticostrone, and LVEDd and increased EF, FS and LVPWT in SS+Ex+ISO in comparison with SS+ISO (P<0.05). Psychosocial stress caused more increase in serum levels of PAB and MDA in rat with heart failure (P<0.001).Application of 4-week exercise caused more improvement in serum levels of PAB and MDA (P<0.001).Conclusion: Psychosocial stress can disturbe functional indexes in heart failure, increases oxidative stress indexes and corticostrone level of serum.Regular exercise could improve some of deleterious effects of psychosocial stress. So, for experiencing more healty situation, both reduction of various stress sources and regular exercise are recommended.

Abstract: 

Background and Objective: Cancers are among the serious problems in communities. In recent years, herbal products have been very promising regarding their anti-tumor and immunomodulatin potentials. SIM5, a herbal preparation, has had good results in discriminating between cancerous cell lines and normal cells, but have not examined directly on patients cells yet. In this study, bone marrow mononuclear cells from cancerous and non-cancerous samples were used to study the effects of SIM5.Materials and Methods: Remained bone marrow samples of multiple myeloma patients (7 cases) and individuals who clinically needed bone marrow examination, but finally were diagnosed with no serious pathologic conditions in their leukocytes (7 cases) were considered as control group. Mononuclear cells were isolated and cultured with SIM5 and R10 fraction. After 48 h incubation, toxicity was assessed using LDH release test and vital activity with MTT test.Results: Cell toxicity with SIM5 was observed only in multiple myeloma samples and not in control group samples. There was a strong correlation between toxicity and the number of plasma cells in bone marrow mononuclear cells. SIM5 induced a decrease in vital activity of cancerous cells but an increase in the normal bone marrow samples.Conclusion: SIM5 has distinct effects on normal and cancerous cells, and its toxic effect is focused on the latter. Therefore, it possesses the properties of an ideal product to be studied more seriously.

Abstract: 

Background and Objective: Heart failure is a clinical syndrome with imminent death in patients and creating stress and anxiety and affected patients could not go to appropriate sleep. In this respect, yoga exercises improve flexibility, muscular power, blood circulation and oxygen absorption and hormone system operation and it causes health feeling. The purpose of this research was determination of effect of yoga exercises on anxiety and sleep quality of patients with chronic heart failure.Materials and Methods: This study was a clinical experiment on 60 patients (2015) divided into two groups of test (30 persons) and control (30 persons).The session of learning and yoga exercises in test group were held two times a week for one hour during two months in heart rehabilitation center in Kerman city under yoga specialist by supervision and approval of a physiologist and a heart specialist. To collect data, Pittsburg sleep quality and Beck anxiety questionnaires were used. Finally, the mean central index was obtained for descriptive statistics and data analysis was done using t test and SPSS software.Results: The findings showed that the mean score of sleep quality and anxiety in test group before intervention was 2.63 and 2.67 and after intervention was 0.79 and 0.87, respectively, that significantly decreased (p<0.001). In addition, average score of sleep quality and anxiety in control group before intervention was 2.43 and 2.37 and after intervention was 2.38 and 2.07, respectively. Also, there was not a significant difference between average score of sleep quality and anxiety in test and control groups before the study. There was also no significant difference between average score of mental quality, sleep delay, sleep disorders before and after yoga exercises in cases with chronic heart failure. But there was a significant difference between average of sleep efficiency, period of sleep, usage of soporific and imperfect operation before and after yoga exercises in in cases with chronic heart failure.Conclusion: The results showed that yoga can be used for empowerment of patients with heart failure in order for heart operation reinforcement and protection of heart abilities. This causes improvement of attitude even with heart failure and improvement of the life style. Finally, it is suggested that these patients take balanced condition after doing this exercise and it is needed for a better life and improvement of heart operation.

Abstract: 

Background and Objective: Ferula szowitsiana is one of the Iranian native medicinal plants that contains variouscomponents such as a pinene, b pinene, limonene, sabenine and myrecene. In several studies, the influence of these components on seizure has been shown. So, in this study the effect of hydroalcoholic extract of Ferula szowitsiana on kindling as a chronic model of seizure in rats was investigated.Materials and Methods: Twenty male rats were categorized into two groups of kindling and kindling-extract (i.p. administration of 400 mg/kg of the extract).For kindling induction, daily intraperitoneal administration of 32 mg/kg of PTZ for 12 days was used. Finally the average of seizure scores was presented as mean±SEM.Results: Ferula szowitsiana hydroalcoholic extract in addition to the significant decrease in severity of seizures in kindling-extract group compared with kindling group (P<0.05), that was visible from day 6, it also delayed kindling process for 2 days.Conclusion: Ferula szowitsiana hydroalcoholic extract is able to reduce the severity of PTZ induced chronic seizure in kindling method. In order to understand its mechanisms in detail, more investigation is needed.

Abstract: 

Background and Objective: Exercise activities have an important role in increasing lean body mass and quality of life in the elderly. Skeletal muscle through the release of myokines have a significant role in muscle growth and improves metabolic condition. The purpose of this study was to investigate the comparison of order of concurrent training on anthropometric characteristics, serum IL-15 levels and insulin resistance index (HOMA) in post-menopausal women.Materials and Methods: Twenty eight healthy post-menopausal women (age: 61.25±0.72 years, height: 156±0.02 cm, weight: 71.70±1.09 kg and BMI: 28.35±0.52) were randomly divided into three groups, strength+endurance (S+E) (n=10), endurance+strength (E+S) (n=9) and control (n=9). Training program was done eight week, 3 times per week. Endurance training were performed on a cycle ergometer (intensity: 61-88 % MHR) and strength training involved selected upper and lower body workout (intensity: 40-75 1RM, 8-18 repeat). Height, weight, BMI, WHR and serum levels of IL-15, glucose and insulin were measured and recorded before and after performing protocol.Univariate (covariance) test was used for analyzing of data.Results: There were no significant differences between experimental groups and control group for insulin (p=0.37). But, there were significant differences in body mass (p=0.02), BMI (p=0.03), fasting glucose (p=0.001), IL-15 (p=0.04) and HOMA (p=0.028). Significant differences were not seen between experimental groups.Conclusion: Concurrent training distinct from order of it, increases muscle growth factor (IL-15) and improves glucose metabolism in premenopausal women, sothese women can take advantage from concurrent training with any order.

Abstract: 

Background and Objective: With regard to the conditions of an emergency patient and the fact that a good emergency health care can save a patient’s life, patient satisfaction levels of emergency health care must be prioritized and the emergency room is the frontline of a hospital’s health care. Despite its importance, few studies in context of factors related to patient satisfaction have been carried out in Iran. The current study has been done with the objective of measuring the level of satisfaction in an emergency health care (in Mostafa Khomeini hospital).Materials and Methods: In this descriptive-analytical study, the satisfaction level of 400 patients with emergency health care was investigated. The patients took part in the study voluntarily. The information was gathered using questionnaires that evaluated the patient’s level of satisfaction. The answers were designed in conformity with Likert scale (great, very good, good, moderate, and bad). This study was approved in Medical Ethics Group and Research Council of Shahed University.Results: In the current study, the patients’ level of satisfaction with the doctors, nurses, nursing services, nursing staff’s attitude, medical staff’s caring attitude, concordance with religious standards, prevention of unnecessary presence of male staff in female patient’s room and also covering the unnecessary parts of a female patient during the examination was reported above average in 100% of the patients and none of the patients evaluated these factors bad. In this study, the amount of costs, comfort care, and the available facilities, the reception staff attitude, laboratory staff, radiology staff and paraclinical services availability were among the least evaluated factors.Conclusion: This study shows a favorable (91.3%) satisfaction in Mostafa Khomeini emergency patients. Medical staff services were evaluated highly satisfactory and the costs, emergency facilities including comfort care and paraclinical services were least evaluated. It seems that improving health insurance conditions, hospital social work, equipping hospitals with comfort care and providing hospitals with dedicated and experienced human resources are points that require most of the emergency managers’ attention.

Abstract: 

Background: Low back pain is one of the most common and most costly chronic diseases, which in many cases there is no cure for it. Iranian Traditional Medicine (TIM), has a valuable therapeutic perspectives for the treatment of low back pain. According to Iranian traditional medicine (TIM), One of the main causes of low back pain is dyspepsia. The aim of this study (by conducting a clinical trial) is to investigate the hypothesis that treatment of dyspepsia can reduce low back pain symptoms.Materials and Methods: 30 patients between 20 and 55 years with chronic low back pain for more than 12 weeks were diagnosed according to the criteria ROM3 suffering from dyspepsia were enrolled.Mentha longifoliacapsules as a drug with positive effect in dyspepsia were given 3 times daily after meals for 4 weeks At the time of entry and after weeks 2, 4 and 8. Visual analog pain scale (VAS) and Oswestry Functional Disability Questionnaire were used and severity of dyspepsia Leeds Questionnaire were checked out.Results: Mentha longifolia capsule significantly reduced dyspepsia, and also pain and disability due to low back pain. (p<0.05). Significant correlation between severity of dyspepsia and pain, and also between dyspepsia and disability from low back pain was observed.Conclusion: Mentha longifolia capsule can reduce pain and disability by reducing dyspepsia. It is suggested to consider digestive problems, especially dyspepsia in patients with low back pain.

Abstract: 

Background and Objective: Diabetes causes damage and impaired renal function. It also causes impaired secretion of endocrine glands that leads to secondary metabolic disorders. Due to the role of curcumin in lowering blood glucose and adjusted antioxidant system, this study was conducted with the aim of investigating the effect of curcumin on the hormones of pituitary-adrenal axis and renal indices in diabetic rats.Materials and Methods: In this experimental study, 32 male rats were divided into 4 equal groups, control, diabetic control and experimental diabetic treated with curcumin (100 and 200 mg/kg, i.p.). The diabetes in diabetic control and experimental diabetic groups was induced using an intraperitoneal injection of alloxan. Curcumin was intraperitoneally injected into experimental groups for 25 days. At the end of injection, the serum levels of urea, uric acid, creatinine, albumin and ACTH, cortisol, and aldosterone hormones were measured. Kidney sections were prepared and examined by means of light microscope.Results: In comparison with diabetic control group, serum levels of urea, uric acid and creatinine in experimental diabetic groups significantly decreased and albumin significantly increased (p>0.05). Regarding ACTH, cortisol and aldosterone hormones, they significantly decreased (p>0.05). Treatment of diabetic rats with curcumin in a dose-dependent manner improved kidney damaged nephrons.Conclusion: Administration of curcumin dose-dependently increased activity of the pituitary-adrenal axis. Also, functional indices of kidney in diabetic rats was close to normal and is has favorable effects on improvement of kidney tissue damages.
