Evaluating the Combination of the Effect of Scaffold with Placental Extraction on Cardiomyocyte Differentiation of Stem Cells: An Experimental Study

Abstract

 Background and Objectives: Disturbance in the structure and function of heart cells can cause cardiovascular disease (CVD) in patients. Although heart transplantation is one of the treatment strategies, it does not apply to all patients due to the challenges associated with transplant rejection. Tissue engineering is one of the new treatment methods used to treat CVD patients today. In this study, the use of polylactic coglycolic acid (PLGA) and polyhydroxybutyrate (PHB) scaffolds along with placental extraction (PE) on the differentiation of cardiac cells from mesenchymal cells has been investigated. Methods: The Electrospinning method was used to make PLGA-PHB and PLGA-PHB-PE scaffolds. Also, tissue culture polystyrene (TCPS) was used for cell culture as a control group. The physical and biological characteristics of the scaffolds, including adhesion strength, water, and protein absorption, as well as their morphology, were evaluated using an electron microscope. Fat samples in this study were obtained from patients undergoing liposuction. To confirm stem cells, CD34, CD45, CD90, and CD105 markers were evaluated using flow cytometry. On the other hand, the viability of the differentiated cells on the scaffolds was evaluated using the MTT method. Also, the expression of four genes Troponin T, GATA4, MYOD, and α-MHC was evaluated using RT-PCR in differentiated cardiac cells. Results: The expression of Troponin T, GATA4, MYOD, and α-MHC genes was significantly higher in differentiated cells on PLGA-PHB-PE scaffolds compared to PLGA-PHB and TCPS scaffolds (P<0. 05). Also, the percentage of survival, water and protein absorption, and adhesion strength was higher in differentiated cells on the PLGA-PHB-PE scaffold than other cells on the other two scaffolds (P<0. 05). Conclusion: The combination of PE with PLGA-PHB can be effective in improving the biological functions of the scaffold and lead to the differentiation of cardiac cells from mesenchymal cells.
