EFFECTS OF SEQUENTIAL MEDIA CONTAINING DIFFERENT CONCENTRATIONS OF GLUCOSE AND PHOSPHATE ON THE ONE-CELL MOUSE EMBRYO DEVELOPMENT

Abstract

 In order to support development of zygotes to the viable blastocysts, one needs to use more than one culture medium, because the metabolite requirements of the preimplantation embryos changes as development proceeds from the zygote to the blastocyst stage. Therefore, this study was initiated to investigate the importance of the GLUCOSE and PHOSPHATE in mouse embryo development by sequential culturing of embryos. Mouse embryos (NMRI) were cultured from one-cell stage (24-26h post-hCG) in different concentrations of GLUCOSE (G0=0, G1=0.2, G2=5.55 mM) in the absence (P0=0) and presence (P1=0.39 mM) of inorganic PHOSPHATE, designated as G0P0, G0P1, G1P0, G1P1,G2P1, and G2P1. All media were supplemented by EDTA (0.02 mM) and BSA (4 mg/ml). At 48h post-culture, some embryos were transferred to renewed media of former one or transferred to G2P0-or G2P1-, supplemented with only BSA (4 mg/ml) -and no EDT A. After 120h, comparison of HATCHING BLASTOCYSTs rate in control and renewal treatments did not show any significant differences (control: 59-71 % and renewal 57-70%). However, hatching rates were increased by sequential transferring from G0P0 G0P1and G1P1to the G2P0-(80, 85, 90%) or G2P1-(79, 90, 74%) when compared to control (59, 69, 71%) and renewed (57, 70, 58%) media. These results indicate that HATCHING BLASTOCYST rate can be increased by SEQUENTIAL MEDIA compared to control or renewal media.
