Abstract

LIPOPOLYSACCHARIDE (LPS) is one of the major components of outer membrane of gram-negative bacteria. It is commonly needed to extract arid purify LPS for Use in research and diagnosis. In this study, we describe a simple and practical method for PURIFICATION of LPS from BRUCELLA abortus S19 strain and BRUCELLA melitensis, 16M strain. This method consists of two stages; first, LPS extracted by aquous butanol and treated by proteinase K. Second, in order to get more purified product, LPS is run on Sepharose CL-6B column or is treated with nucleases (RNase and DNase). The degree of purity of LPS was determined by chemical analysis and electrophoresis in sodium dodecylsulfate, polyacrylamide gels. The results show that: 1) protein and nucleic acid impurities of LPS are respectively 1 and <0.5 percent following gel filtration and 2.5:and <0.2 percent following nucleases treatment; 2) Electrophoretic pattern of LPS consists of a fast-migrating band representing the lipid A and core oligosaccharides and slowly migrating components appear as a smear, representing the intact LPS.
