EFFECT OF ANTI-INFLAMMATORY DRUG (PIROXICAM) ON THE DEVELOPMENT OF FATTY STREAK AND BIOCHEMICAL FACTORS IN CHOLESTEROL-FED RABBITS

Abstract

 Background and aims: CARDIOVASCULAR DISEASES are the most prevalent cause of mortality all over the world. Efforts for CARDIOVASCULAR DISEASES prevention and treatment must be directed to their underlying mechanisms. One of the mechanisms is INFLAMMATION. The effect of the anti-inflammatory drug, PIROXICAM, on the development of FATTY STREAKs has been investigated in this study. Methods: Twenty male rabbits were randomly divided into four groups fed with four types of regimen: a) normal rabbit chow, b) 1% cholesterol diet, c) cholesterol diet plus PIROXICAM 6mg daily, and d) normal rabbit chow plus PIROXICAM. The amounts of cholesterol, LDL, HDL, triglyceride and fasting blood sugar (FBS), C-reactive protein (CRP), malondialdehyde and antioxidant capacity were determined at the beginning and the end of the study. After 12 weeks, the animals were sacrificed and the aorta, as well as the right and left branches of the coronary artery were dissected, and histological processing was carried out. Results: The results indicated that the mean grade of FATTY STREAKs of right and left coronary arteries as well as aorta were significantly different between normal diet and cholesterol diet groups (P<0.0001). PIROXICAM supplemented with cholesterol significantly decreased progression of FATTY STREAK in the right and left coronary arteries (P<0.05) comparing with the high cholesterol diet group. Conclusion: This study showed that PIROXICAM decreases progression of FATTY STREAK by reducing CRP, LDL, TG and increasing antioxidant capacity and HDL in rabbit suggesting that PIROXICAM, has beneficial effects on preventing the development of FATTY STREAKs.
