Abstract

 Introduction. Lung cancer is the most common visceral malignancy in men. The incidence is increasing dramatically in women, and lung cancer has passed breast carcinoma as a cause of cancer death in women. Overall lung cancer is the most fatal malignancy. TP53 is a well-studied tumor-suppressor gene and it is thought to play a role in the control of the cell cycle. The wild type TP53 protein inhibits cell proliferation by arresting cells in the G1 phase of cell cycle, and loss of this activity can lead to neoplastic transformation. A little over 50% of human tumor contain mutations in this gene. In lung cancer, the nodal involvement is the best prognostic factor. This study was conducted to determine whether immunohistochemical analysis of TP53 in patients with non - small - cell lung cancer (NSCLC) in associated with nodal involvement. Methods. This retrospective cross-sectional study, carried out on 120 cases of primary lung carcinoma. From each-paraffin block two slides were prepared. one with H&E staining and another with TP53 staining. Data were analyzed by using Chi-square test.Results. Tumor samples from 120 patients with NSCLC were studied. Positive nuclear staining was detected in 69(57.5%) tumors. 30.69(43.4%) were negative for lymph node involvement whereas 39.69(56.5%) were positive. Statistically significant correlation was not found between TP53 expression and nodal involvement. Also, there was no significant correlation between the histopathological subtype of NSCLC with TP53 mutation and nodal involvement.Discussion. Because, the results would be easily influenced by various factors, such as the methods used to detect TP53 mutations and the characteristics of study populations, the latter of which include tumor histology and staging, as well as ethnic group, sex, smoking habit, geographic conditions, We believe that is important to compare different measuring methods used to detect TP53 mutations and that further studies on the patients characteristics associated with TP53 mutations should be conducted.
