Abstract

 Introduction: The factors and intercellular relationships controlling the proliferation of the stem cells and their differentiation into different committed Precursor cells are of fundamental importance in understanding the mechanisms controlling hematopoiesis. Based on the model proposed by Ogawa in 1989 hematopoietic proliferation is controlled by a cascade of growth factors like erythropoietin ,Interleukin 5 and G - CSF. In This study the effects of Interleukin 2 and 5 and IFN-y and combination of IL-2 & IL-5 on the development of murine hematopoietic cells was investigated.Methods: Bone marrow cells were extracted from tibia and femur of Balb/c mice and cultured in complete culture medium (IMDM containing 12.5%FCS 12.5%HS I micro M, Hydrochortison) on microplates at 33.c with 5%C02. Then Interleukin 2 and 5 were added to the cells separately and in combination. The colony assay way performed weekly for five weeks.Results and Discussion: The results showed that Interleukin 2 decreased the number of CFU-GM hematopoietic colonies and combined with Interleukin 5 affected the development of murin hematopoiesis and increased the number of CFU-GM colonies.
