Abstract

 Introduction: Hydatidosis is one of the most important zoonosis. Because of the importance of this disease and also because of the effects of strain variation on strategic planning for the control of this disease, animal and human hydatid cyst isolates were evaluated for strain variation.Methods: Fifty five animal hydatid cyst isolates and 15 human isolates were collected from slaughterhouses and hospitals of Isfahan. Total genomic DNA were extracted from hydatid cyst protoscoleces using method of SDS and proteinase K treatment, phenol/chloroform extraction and ethanol precipitation. rDNA - ITS1 region of all isolates were amplified using 2 specific primers (BD1 and 4S) and the PCR products were electrophoresed and stained.Results: rDNA - ITS1l fragment size of all sheep, goat, most of the cattle, human and some of the camel isolates were the same size about ~ 1000 bp. Wherease that fragment in most camel, some cattle and human isolates had the same pattern about ~1100 bp.Discussion: Based on rDNA - ITS 1 fragment patterns in the animal and human isolates, it can resulted that at least two strains of E. granulosus exist in Isfahan. The sheep strain (G1) is dominant genotype in this region and the camel strain (G6) that is also present in the region, can infect human the same as sheep strain.
