Abstract

Introduction. Escherichia coli 0157:H7 is an important cause of Hemorrhagic Colitis, Hemolytic-Uremic Syndrome and Acute Renal Failure. In order to recognize this bacteria 750 stool samples from diarrheal patients that referred to Al-zahra Hospital examined by PCR and bacteriological methods.Methods. In bacteriological methods all samples cultured on SMAC medium and then sorbitol negative colonies were isolated and examined by Biochemical tests and E.coli 0157:H7 latex reagent. Multiplex PCR technique performed on all of the samples by 3 serios perimer stxl, stx2, rfb.Results. In bacteriological exams 11 samples from the 750 sample were positive. In PCR method 29 samples were verotoxigenic E.coli that 14 sample were relative to 0157:H7 and 15 samples relative to Non-0157: H7 E.coli. Sensitivity and specifity of PCR method that we use incontrast with culture respecitvly is 99% and 100%.Discussion. With the respect of present study results that PCR method known as a sensitive and reliable methods and urposed as a suitable method for recognition of Escherichia coli 0157:H7 on clinical, food samples and epidemiological study.
