Abstract

 Objective: BREAST CANCER is multidimensional disease in initiating trends inflicting almost every aspect of cell cycle. METASTASIS (chain of non-randomized sets of events) is usually influenced by abnormal inflictions induced either at genetic or epigenetic levels. Current objective of this study is to identify and characterize sets of those molecules which are found responsible for suppression of METASTASIS cascade. Our earlier research highlighted the putative role of CD82 (a tetraspanin family member) in suppression of BREAST CANCER METASTASIS progression. A central dogma based analysis of this molecule (germline mutation and expression screening) in BREAST CANCER affected patients highlighted its potential significance in disease progression as well as overall patient’s survival (p=0.04) For a thorough insight, effect of this molecule on cancer cells proliferation, adhesion, motility and invasion had also been studied. Materials and Methods: Transgenes based cancer cell lines (both forced expression in MCF-7 as well as knock out in MDA-MB-231) were generated. Results: After extensive experimentation, a significant correlation of CD82 along with cancer cell adhesion (p=0.021), cellular invasion (p=0.022) and motility (p=0.024). CD82 did not impart any significant role on cancer cells growth.Conclusion: From this data it has been evidential CD82 substantially contribute to cancer cells regulation in metastatic cascade. Another elucidated sphere of this study is to extract and characterize such sets of molecules that impart a substantial role in this METASTASIS process in relation to BREAST CANCER. Identifying and multiplexing those markers along with their potential role will help in designing a tool for early prognosis. METASTASIS suppressor genes may be also be set a universal set of marker for tracking METASTASIS initiation, promotion and progression.
