Title

Evaluation of Melatonin on HSP60 and HSP70 Gene Expression Following High-Intensity Anaerobic Training after Ischemia-Reperfusion in Male Rats

Abstract

 Background and Objective: The aim of the present study was to evaluate the effect of Melatonin supplementation on HSP60 and HSP70 gene expression changes following high intensity anaerobic exercise in male rats after Ischemia-Reperfusion. Materials and Methods: Twenty four male Wistar rats weighing approximately 200-250g were divided into 4 groups of 6 each: study groups including control, Melatonin, Anaerobic Training, and anaerobic+Melatonin training. The anaerobic exercise protocol was performed for one month, while the Melatonin Anaerobic Training group underwent Melatonin gavage during the exercise. At the end of one month, rats were injected with isoprenaline after two days of rest to induce Ischemia-Reperfusion. After two days the rats were killed, their left ventricle removed and the expression levels of HSP60 and HSP70 genes were measured by RNA extraction and Real Time technique. Results: Results showed that Melatonin consumption decreased HSP60 and increased HSP70 apoptotic gene expression after anaerobic exercise (P <0. 05). Conclusion: The results of this study showed that intake of Melatonin concurrent with intensive anaerobic exercise reduce the volume of myocardial apoptosis after Ischemia-Reperfusion-induced Ischemia-Reperfusion.
