STUDY OF CYTOTOXIC EFFECTS OF VEROTOXIN1 AND MONOPHOSPHORYL LIPID A ON MCF-7 CELL CULTURE

Abstract

 Introduction: Verotoxins are subunit toxins, that involved in pathogenesis of enterohemorrhagic Escherichia coli. Early studies of cytotoxicity on tumor cells and some human cancers indicated that VEROTOXIN1 effective in (in vitro) and laboratory animals. Because of specific receptors on studied tumor cells, VEROTOXIN1 exerts selective cytotoxicity.Our purpose is purification of VEROTOXIN1 from verotoxigenic E.coli, to study cytotoxic effects of VEROTOXIN1 and Monophosphoryl lipidA (MPL) on MCF-7 cells.Materials & Methods: Toxin production in five verotoxigenic strains of Escherichia coli was screened by Verotoxin-producing E.coli-Reverse passive latex agglutination test (VTEC-RPLA kit ,Oxoid,Co,Ltd) and then purified by affinity chromathography of receptor glycolipid (P1 blood group antigen). Then MCF-7 cells treated with serial dilutions of VEROTOXIN1 and MPL in separate and combined forms. Viability (Dye exclusion method) and fragmentation of treated cells was studied.Findings: Our results indicate that MCF-7 cells have susceptibility to VEROTOXIN1, and 33 nanograms of toxin per Milliliter have cytotoxic dose 50% on treated cells. And DNA fragmentation studies indicate evidences of apoptosis. Whereas MPL is a non toxic derivative of lipopolysaccharide that have synergistic activity on cytotoxicity of VEROTOXIN1. Conclusion: Antitumoral effects of VEROTOXIN1 have been shown for several years, but scientists interesting on discovery of agents that have synergistic effects with known antitumorals. MPL is one of the microbial metabolites that we report its synergistic antitumoral effect with VEROTOXIN1 against MCF-7 cells.
