Abstract: 

BACKGROUND: Antimicrobial peptides with relative length (2-100 amino acids) and positive charge (pure charge (+9)-(+2)) are amphiphilic that isolated from a wide range of animals. Recently, these peptides have been known as a part of innate immune response. Nowadays, more than 500 antimicrobial peptides from animals have been reported.OBJECTIVES: The aim of this study was to evaluate the anti-Candida effects of cationic peptides derived from Rana ridibunda skin.METHODS: In this study, using alcohol-acid technique, peptides of frog’s skin were isolated and purified by Sep-Pack and Sephadex column. Then the anti-Candida activity (Fluconazole Resistance C. albicans, Fluconazole Sensitive C. albicans, and C. glabrata) of the peptides in different concentrations were evaluated.RESULTS: Regarding to statistical analysis, peptides in concentration ranging from 25 to 100 mg/ml had the most anti-Candida activities. In respect to different understudy Candida species, these agents had the less effect on the Fluconazole Resistance C. albicans (p<0.05).CONCLUSIONS: The anti-Candida effects of cationic peptide obtained from frog skin are approved in this study.

Abstract: 

BACKGROUND: Vibrio paraheamoloyticus is one the causative agents of vibriosis with high mortality in farmed fish and shrimp and under predisposing conditions.OBJECTIVES: This study was aimed to assess the effect of gamma irradiation on the inactivation of V. paraheamoloyticus under fresh and freeze-dried conditions.METHODS: Vials of 0.5 ml fresh culture of the bacterial suspensions at 1.5*1010 cfu/ml were subjected to 1, 1.5, 2, 2.5, 3 and 4 KGy. Also, vials of 0.5 ml freeze-dried culture of the bacterial suspensions at 1.5 x 1010 cfu/ml were subjected to 2, 4, 7 and 8 KGy. The bacterial growth behavior was then evaluated on fresh medium.RESULTS: The obtained results showed that the minimum doses of 4 and 10 KGy were sufficient for the inactivation of fresh and freeze-dried bacteria, respectively.CONCLUSIONS: The result of this study shows that inactivation of V. paraheamolyicus in fresh culture condition requires below half- dosage of gamma ray required for the inactivation of the freeze- dried of bacterial cells.

Abstract: 

BACKGROUND: The use of chemical materials such as malachite green to disinfect fish eggs is banned in many countries due to its reversed environmental effects.OBJECTIVES: The purpose of this study was to investigate the effect of metalonic sumac extract as antifungal agent on eggs of Schizothorax zarudnyi during the incubation period and to compare its effects with malachite green.METHODS: 24 hours after fertilization, eggs were treated with four concentration of Methanolic sumac extrat (500, 750, 1000, 1250 mg/L) using static bath method every 12 h for 20 min and 0.1 mg/L malachite green using the same method every 12 h for 6 min. There was also a control group with no treatment to better analysis.RESULTS: The results showed that the maximum and minimum fungal infection was observed respectively in control group with %20.23±0.51 and the samples treated with 1250 mg/L sumac extraction with %0.91±0.51. Furthermore, no significant differences in fungal infection, fertilization and survival rate were found among treated and untreated eggs (p>0.05).CONCLUSIONS: Since no apparent deformities and abnormalities were observed in hatched larvae, suggested sumac extract with 500 mg/L concentration, as an environmental friendly matter, can be used for disinfection of S. zarudnyi eggs during incubation period.

Abstract: 

BACKGROUND: The hazardous nature of aflatoxins to human and animals necessitate the establishment of control measures.OBJECTIVES: The effect of two medicinal plants, Satureja khozistanica and Satureja macrosiphon, was studied on inhibiting Aspergillus flavus growth and reducing aflatoxin B1-content in the liquid medium.METHODS: Essential oils were isolated by hydrodistillation method, using a Clevenger-type apparatus. Various extracts of plant materials were macerated with various extraction solvents (ethanol, ethanol70% and water extracts). Essential oils (0, 62.5, 125, 250, 375 and 500 mg/l) and various extracts (0, 500, 1000, 2000, 4000 and 6000 mg/l) of S. khozistanica and S. macrosiphon were examined for reducing A. flavus growth and it’s AFB1-content in the liquid medium. Amount of aflatoxinB1 was evaluated by high performance thin layer chromatography method.RESULTS: Essential oil of S. khozistanica at the concentration of 375 mg/l as well as its ethanol and ethanol 70% extracts at 4000 and 6000 mg/l respectively caused complete inhibition of fungus mycelial growth, whereas essential oil and extracts of S. macrosiphon couldn’t inhibit Aspergillus growth completely even at the maximum concentration. Essential oils of S. khozistanica and S. macrosiphonia at the concentration of 250 mg/l reduced AFB1-production 98 and 33.52% respectively. Various Extracts of S. khozistanica exhibited stronger anti-AFB1-biosyntesis activity than those of S. macrosiphon, so that, ethanol, ehanol70% and aqueous extracts of S. khozistanica at 4000 mg/l reduced 100, 96 and 32.37% of AFB1-production, respectively. On the contrary, essential oils, ethanol and ehanol70% extracts of both plants couldn’t significantly degrade AFB1-contamination, whereas aqueous extractsof S. khozistanica and S. macrosiphonia at the concentration of 4000 mg/l resulted in degradation of 25 and 32.16% AFB1-content, respectively.CONCLUSIONS: In general, Essential oil and ethanol extract of S. khozistanica considerably inhibited A. flavus growth as well as AFB1-biosynthesis while aqueous extract of S. macrosiphon showed strong AFB1-degradation activity.

Abstract: 

BACKGROUND: H9N2 avian influenza viruses (AIV) A have become panzootic in Eurasia over the last decade and have caused several human infections in Iran since 1998 and inactivated vaccine has been used in chickens to control the disease.OBJECTIVES: The purpose of this study was to analyze H9N2 viruses that have infected broiler in Tehran Province, Iran between 1998 and 2008 based on Matrix gene.METHODS: The complete coding region of Matrix (M) gene from 8 of H9N2 subtype isolated from chicken flocks in Tehran Province during 1998-2007 was amplified and sequenced.RESULTS: Sequence analysis and phylogenetic studies of H9N2 viruses on the basis of data of viruses in this study and other selected strains available in the GenBank were conducted and determined variations among these sequences at different levels. Sequence analysis revealed a large number of similar substitution mutations and close evolutionary relation among sequences of M gene. Phylogenetic analysis showed that all our isolates belonged to the G1-like sublineage. In this study, it was determined that Iran’s isolates have been in two separate branches and have the most similarity with Pakistan, United Arab Emirate and occupied Palestine’s isolates.CONCLUSIONS: The available evidence indicates that M genes of H9N2 circulating in Iran during the past years were not well conserved. Our finding emphasizes the importance of reinforcing AIV surveillance.

Abstract: 

BACKGROUND: Granuloca cells have a key role during estroeidogenesis.OBJECTIVES: The purpose of this study was to determine the effect of number of culture medium granulosa cells on estradiol concentrations and mRNA codding estrogenic and progestagenic enzyme.METHODS: Briefly, follicles between 2 and 5 mm diameter were dissected from the ovaries of adult cows and were collected by rinsing the follicle walls with Dulbecco Modified Eagle medium/F12 (DMEM/F12). The number of cells was counted with a haemocytometer and the viable cells were assessed by the dye exclusion method using 0.4% Trypan Blue. Treatments were 1) 500,000 cell/500 ml, 2) 250,000 cell/500 ml, 3) 500,000 cell/200 ml 4) 250,000 cell/ 200 ml. All data were analyzed by JMP (SAS).RESULTS: Low plating density increased E2 secretion and mRNA encoding LHR, FSHR and estrogenic enzymes (17bHSD, CYP19), whereas decreased mRNA encoding GADD45b. There were no differences among treatments for RNA and protein concentration. Low plating density also decreased protein amount but there was no difference among treatments for RNA amount. In conclusion, decreased cell density cause increase in mRNA encoding codding estrogenic enzyme gene expression and decrease in mRNA encoding progestagenic enzyme gene expression.CONCLUSIONS: Protein concentrations did not changed with decreased cell density therefore we can save cells against harmful effect of increasing cell density.

Abstract: 

BACKGROUND: Major histocompatibility complex (MHC) plays a central role in regulation and control of the immune responses to infectious diseases. Due to its polymorphism, individual differences in response to vaccines have been observed in different chicken populations. Studying the association of chicken MHC with immune response to vaccines will help the control of infectious disease and vaccination success.OBJECTIVES: The present study aimed to evaluate the MHC polymorphism and its association with antibody response against infectious bursal disease (Gumboro), Newcastle (ND) and Influenza (AI) vaccines in Khorasan native chickens.METHODS: Diversity of LEI0258 microsatellite marker (MHC genotyping) was investigated by fragment analysis method. Antibody titer against IBD was measured by ELISA and antibody titers against ND and AI vaccines were measured by Haemaglutination Inhibition (HI) assay. Statistical analysis was performed using SPSS software (version 21). Univariate regression analysis was performed using weighted least squares with weight number of progeny mean data.RESULTS: Total of 13 LEI0258 microsatellite alleles were identified in Khorasan native chickens which indicated a high genetic diversity in the population. The allele 361 bp had the highest (28.48%) and the allele 350 bp had the lowest (0.69%) frequency, respectively. In evaluating the association of MHC with immune responses, 311 and 313 bp alleles were significantly associated with elevated immune responses to Newcastle vaccine, while allele 266 bp was associated with lower IBDV antibody titers (p<0.05).CONCLUSIONS: According to the important role of MHC in controlling infectious disease resistance or susceptibility and quality of immune responses, these results could be used for selection and improving the populations under selective breeding.

Abstract: 

BACKGROUND: Application of different sources of fatty acids and their effect on ovarian performance is a current issue in animal science and research centers.OBJECTIVES: The purpose of this study was to determine the effect of different fatty acids in close-up period on population and size of follicles and measuring blood parameters related to the energy state of the body after calving of lactating dairy cows.METHODS: In this study 15 Holstein cows were selected by expected date of parturition (around 30 days before calving) and assigned them randomly within treatments diets. The diets were formulated to be isonitrogenous. Cows were fed 1) control (carbohydrate source), 2) Rumen-protected fat (RF) (saturated fatty acids) and 3) Roasted Soybean (RS) (unsaturated fatty acids). Blood sample was collected at 7 day intervals in -21, -14, -7, 1, 7, 14, 21 periods of calving. Also dry matter intake and body condition score cows were recorded. Ovarian activity was monitored by daily transrectal ultrasound scanning from 30 to 40 and 61 to 70 DIM.RESULTS: Average plasma glucose and cholesterol concentration were significantly different among treatment groups. Numerically, follicular classes were higher in cows, fed with fat diet compared with control group.CONCLUSIONS: According to the results of this experiment, it seems that application of polyunsaturated fatty acids, to improve the energy and ultimately the function of the ovaries, has more advantage than saturated fatty acids.

Abstract: 

BACKGROUND: Minerals are essential materials in animal reproduction. The determination of these elements is valuable in relation to different conditions in Arabian horses.OBJECTIVES: The purpose of this study was to evaluate the effects of pregnancy on certain macro and micro minerals in Arabian horses.METHODS: Blood samples were collected from one hundred apparently healthy mares from different regions of Ahvaz including 50 pregnant mares (light, medium, heavy) and 50 non-pregnant mares. Macro-minerals (calcium, phosphorus, magnesium, sodium and potassium) and micro-minerals (copper, zinc and iron) were measured, using standard common laboratory methods. Methods of O-cresol phetalein, Molybdate reduction, Xylidil blue were used for measurement of calcium, phosphorus and magnesium respectively. Flame photometry was used for measurement of sodium and potassium. Amounts of copper, zinc and iron were measured by atomic absorption spectroscopy.RESULTS: In order to analyze the results, all the data was statistically compared by SPSS-17.0 software. The study of serum calcium levels between the pregnant and non-pregnant mares showed that the differences were not significant. In this study there was significant differences between serum phosphorus levels between pregnant and non-pregnant mares (p<0.05). The serum phosphorus levels during different stages of pregnancy was significantly different (p<0.05), and the level of serum phosphorus during pregnancy declined in total compared to non-pregnant mares. Serum magnesium did not show significant differences between two groups. Serum sodium levels, compared to the other two groups, did not show significant changes. Changes in serum potassium levels showed significant difference between two groups of pregnant and non-pregnant mares (p<0.05). The level of potassium in pregnant mare’s serum was less than non-pregnant, and the least level of this element was seen in medium pregnancy.CONCLUSIONS: Except phosphorus, potassium and zinc, other minerals did not affected different stages of pregnancy. Although the relationship between these changes and pregnancy can be the result of physiological processes, the other factors such as parity, age, nutrition, season, weather conditions and other factors should not be ignored in the evaluation.

Abstract: 

BACKGROUND: Brownbanded Bamboo sharks (Chiloscyllium punctatum) is one of the cartilaginous fish that lives in the Persian Gulf.OBJECTVES: The purpose of the present study is to study the effects of thyroid hormone on reproductive system of this specious.METHODS: In this investigation, 36 individual of Brownbanded Bamboo sharks (Chiloscyllium punctatum) in both autumn (before reproduction) and spring (period of reproduction) seasons were caught from Dervish’s Creek located at the northern of Persian Gulf. After weighing, biometry, sex determination and the numbering of sharks, (the sex is determined through clasper which is observed in male and not in female), ELISA was used to measure serum thyroid hormone concentrations.RESULTS: The results showed that T3 and T4 hormones were increased dramatically in the reproductive season compared with the non-breeding season (p<0.05). On the other hand, by classifying the sharks into different groups, we found that the levels of thyroid hormone were higher in the blood of larger sharks compared with the smaller ones (p<0.05).CONCLUSIONS: It is probable that T4 hormone levels will increase dramatically for the development of eggs in the female shark.

BACKGROUND: Essential oils as growth stimulant play an important role in improving intestinal microflora and morphological properties and immune system in broiler chickens.OBJECTIVES: The aim of this study was to investigate the effects of Artemisia sieberi oil on intestinal morphology, secum microflora and immune system in broiler chickens.METHODS: Two hundred day-old broiler chickens (Ross 308) were allocated to 5 treatments, 4 replications with a completely randomized design during 42 days of age. Experimental treatments consisted of control (basal diet) or basal diets containing different levels of Artemisia sieberi oil (100, 200 and 300 mg/kg) and 600 mg/kg flavophospholipol antibiotic. On day 42 days of age, intestinal morphology and microflora population and immune system were evaluated by measuring the weight of bursa of fabricious and spleen and antibody production.RESULTS: The results indicated that the highest lactobacillus count and the lowest Escherichia coli count of the caecum was found by inclusion of 300 mg/kg Artemisia sieberi oil in the diet (p<0.0001). The antibiotic and Artemisia sieberi oil treatments showed higher villus height in the duodenum compared with control group (p<0.01). Artemisia sieberi oil and antibiotic supplementations significantly decreased epithelial thickness and goblet cell number of the small intestinal compared with control group (p<0.05). The dietary supplementation did not significantly affect the crypt depth and villus height to crypt depth ratio in small intestine. The level of 300 mg/kg Artemisia sieberi oil significantly increased antibody titration against Newcastle disease virus (p<0.01).CONCLUSIONS: Adding Artemisia sieberi oil at levels of 300 mg/kg to broiler chicken diets can improve gut microflora (as measured by changes in populations of Escherichia coli and lactobacillus).

Abstract: 

BACKGROUND: Maturity of silver carp (Hypophthalmichthys molitrix) ovary takes place at 2-8 years old and it is depends on photoperiod and environmental water temperature.OBJECTIVES: The aim of this study was to identify macroscopical and microscopical structure of silver carp ovary in two categories of body weight, total body length and at 2 and 4 years old in Khuzestan province climatic conditions.METHODS: 20 silver carp ovaries were studied in two groups: Group 1(G1): 10 fish with total body length 46±1.33 Cm and weight 1050±0.09 grand approximately 2 years old. Group 2(G1): 10 fish with total body length 86±3.64 Cm and weight 5460±800 gr and approximately 4 years old. For the histometrical studies, 5-6 m sections were made by routine paraffin embedding method and stained by H&E and PAS.RESULTS: Microscopic results showed that ovaries of G1 covered by a thin loose connective capsule which numerous of egg-bearingtrabecula extended from capsule into ovaries. These trabecula contained primary germ cells, oogonium, chromatin nocleolus, perinocleolus and cortical alveolus follicles. Ovaries of G1 do not have yolk and mature follicles. But in ovaries of G2, growth and mature follicles were observed and mature follicles were most follicles. Histometrical results showed that there are not a significant differences in diameter of follicles, oocytes and oocytes nucleolus in G1 and 2. The maximum and minimum diameter of follicles were seen in mature and chromatin nucleolus follicles respectively. The Zona plucida thickness was increased from corticalalveolus (1.53±1.11 mm) to mature follicles (6.88±0.21 mm) significantly (p<0.05).CONCLUSIONS: The most interesting finding of this study is that in Khuzestan Province climate, the ovary of silver carp fish in G1, with average weight 1050±0.09 gr and total length 46±1.33 Cm and approximately of 2 years old is immature but ovary of G2 with average weight of 5460±800 gr and total length 86±3.64 Cm and approximately 4 years old is mature.

Abstract: 

BACKGROUND: Identifying the potential of livestock and forage plants of each region is a way to compensate the lack of forage, and efficient use of edible resources of the country. Milk Thistle is a medicinal plant which may be used as fodder for native livestock of Khuzestan Province.OBJECTIVES: The purpose of this research was to study the effects of different levels of Milk Thistle on rumen fungi and whole rumen microorganisms (WRM) of Khuzestan buffalo, with diets containing grains with different degradation rate (barley and maize).METHODS: The impact of diets containing different levels of Milk Thistle (0, 100 and 200 g per kg DM) on rumen fungi and WRM of buffalo was measured by different techniques.RESULTS: Potential and rate of gas production from experimental diets by WRM were not significantly different. This parameters by buffalo rumen fungi were significantly differed and increased in diets containing Milk Thistle (p<0.05). Adding Milk Thistle in the barley-based diet increased dry matter and NDF digestibility numerically while in the corn-based diet dry matter and NDF digestibility was slightly reduced. In the SRFCM, NDF digestibility, in both basal diets in 9 days and during total period was affected by diets containing Milk Thistle and decreased NDF digestibility with increasing levels of Milk Thistle (p<0.05). Regardless of the type of basal diet, digestibility of NDF (p<0.05) and dry matter (p>0.05) for day 6, were increased with increasing the amount of Milk Thistle.CONCLUSIONS: In general, the use of Milk Thistle did not have negative effect on microorganisms and digestion of nutrients by them. Therefore, results suggest that Milk Thistle could be used up to 20 percentage in buffalo diet without any negative effect on digestion and fermentation characteristics by total microorganisms and fungi.

Abstract: 

BACKGROUND: Inclusion of antioxidant sources in feed or water reduces the detrimental effects of free radicals and increases the antioxidative potential and stability of lipids in heat stressed broiler chicken.OBJECTIVES: This study was conducted to examine the effect of Satureja khuzistanica essential oils (SkEO) on early postmortem and ultimate pH as well as antioxidative potential of breast muscle in broiler chicken.METHODS: A total number of 720,1-d-old Arian broiler chicks were used in a 42-d trial to examine the effects of six experimental treatments consisting inclusion of 0 (control-), 200, 300, 400, 500 mg/L SkEO and 500 mg/L Polysorbate-80 (control+) in drinking water in six replicates of 20 birds each in a completely randomized blocks design. Catalase enzyme activity (by kinetic method) and glutathione peroxidase as well as superoxide dismutase activities (via Randox® kits) were measured spectrophotometerically. Lipid peroxidation also was measured in breast muscle by a chemical method with thiobarbituric acid.RESULTS: The breast percentage and early as well as ultimate pH of breast muscle was not differ for the birds received SkEO-treated water compared to the control birds (p>0.05). Addition of SkEO in drinking water at doses greater than 200 mg/L significantly decreased the thiobarbituric acid reactive substances values in breast muscle of the birds in comparison with the control birds (p<0.05). Catalase activity was significantly higher in breast muscle of the SkEO-treated groups than the control groups (p<0.05). Activity of glutathione peroxidase was significantly greater in the birds given 400 and 500 mg/L SkEO. Addition of SkEO into water up to 400 mg/L increased superoxide dismutase activity in breast muscle of the treated birds compared to the control birds.CONCLUSIONS: It was concluded that supplementation of drinking water with SkEO elevates the antioxidative potential and increases the lipid stability of breast muscle in heat stressed male broiler chicken.

Abstract: 

Neuroendocrine cells are of the peptide and amine secreting cells and because of endodermal origin, exist in respiratory and gastrointestinal system. The report confirms the presence of a tumor using immunohistochemical methods. A hepatic mass was detected in a 10-year-old male mixed-breed dog which was referred with clinical history of frequent vomiting, anorexia, lethargy and syncope. Biochemical and hematological abnormalities included elevation of ALT and AST levels, leukocytosis and anemia. Radiographically the large hepatic mass and multifocal metastatic nodular areas in the other organs were observed. At necropsy the liver had a multilobulated firm mass involving whole caudate lobe of the liver. On the cut section, the tumor was grey sanguineous with hemorrhagic surface and numerous necrotic foci. Microscopically, there were unencapsulated, highly cellular neoplastic proliferations with few hemorrhagic foci in the liver that had a trabecular pattern to Rosette formation which were separated by a fibrovascularstroma. The neoplastic cells were round to oval, with granular eosinophilic cytoplasm and hyperchromatic nuclei. Metastases were observed in lungs, kidneys, mesenteric lymph nodes and even in the arteries of the tunica albuginae of the testes. Immunohistochemically, neoplastic cells were immunoreactive for NSE and S100 protein. They were negative for CEA, Chromogranine A, CD34, AE1/AE3, CK20 and Hepatic Antigen (HA). Negative CEA, CK20, CD34 and HA disproved the probability for collangiocarcinoma, metastatic carcinoma, hematopoietic and vascular origin and hepatocellular carcinoma of this tumor respectively. Immunohistochemical demonstration of NSE and S100 protein supported the diagnosis of the Carcinoid like. However, negative reaction for Chromogranine A does not disprove neuroendocrine carcinoma diagnosis.
