Abstract: 

Introduction: Sirtuin 1 is a protein deacetylase that plays an important role in many cellular processes. The serum sirtuin 1 levels are higher in the polycystic ovary syndrome (PCOS) patients than the healthy subjects. The aim of this study was to assess the role of SIRT-1 rs7895833 polymorphism in the susceptibility of PCOS.Methods: This case-control study was done on 456 Iranian women from March to February 2015. Blood samples were collected from 274 women with PCOS and 182 age matched (±5) healthy women at Shiraz Ghadir Mother and Child Hospital. After DNA extraction, SIRT1 rs7895833 genotype determination was done by Tetra-ARMS PCR. Data analysis was performed by SPSS version 18.Results: In the dominant model for G allele (AG+GG vs. AA), AG+GG genotypes in SIRT1 rs7895833 gene polymorphism was associated with the increased risk of PCOS (OR: 2.01, %95CI:1.32-3.24, P=0.002). Moreover, there was a significant association between the G allele and PCOS (OR: 1.45, %95CI:1.10-1.91، P=0.008).Conclusion: The results showed a possible association between SIRT1 rs7895833 gene polymorphism and susceptibility to PCOS.

Abstract: 

Introduction: Depression prevalence during pregnancy and its prevention and treatment has been concerned by researchers. The aim of study was to investigate the effects of physical activities in this period on post-term developmental indexes.Methods: The research method was quasi-experimental one. 3 groups with pretest and posttest design with control group [depressed (n=27), depressed with physical activities (n=32) and the control (n=30) groups] were studied. Beck depression questionnaire was used to measure the rate of depression. Aerobic training program was 3 sessions a week, each session 30 min by 40-50% of maximal reserve heart rate. Infants’ height, weight and head circumference measurements were obtained after birth. One way ANOVA and Tukey post-hoc test were used for statistical analysis.Results: The results indicated that height, weight and head circumferences as three infants’ growth indexes were lower in depressed group compared to the other groups (p<0.05). Control group and depressed mother with physical activities gave a birth with the better developmental indices, respectively. Another results showed that depression level in the depressed group with physical activities was improved from high intensity to medium one (p=0.001).Conclusion: Conclusively، regular physical activity plays a key role in depression improvement of pregnant women and it leads to the improvement in infants’ growth indexes.

Abstract: 

Background and Objectives: One of the ways to strengthen the effect of vaccines is using the adjuvant. STxB has a carrier and adjuvant role that we can fuse it with vaccine candidate antigens and produce efficient vaccines. BLF1 has a role in the pathogenesis and infection by Burkholderia pseudomallei, that can be studied when fused with stxB.The aim of this study expression blf1-stxB gene Cassette in E. coli and antibody production in mice.Methods:In this study the synthetic gene in pUC57 plasmid was purchased from Nedaye Fan COR. pUC57 plasmid containing blf1 with BamHI, SalI restriction enzyme sites was subcloned in pET28a(+)-stxB expression vector and transformed into E. coli BL21 DE3. expression blf1-stxB gene Cassette was induced by IPTG. After extraction by affinity chromatography, the recombinant protein was injected four times to mice.Results: In this study, blf1-StxB cloned gene in pET28a(+) expression vector was approved by PCR and enzymatic analysis. Also recombinant protein confirmed by SDS-PAGE and Western blotting. Then antibody produced from the mice serum, were isolated and confirmed by ELISA.Conclusion: Given that BLF1 protein has the ability to stop protein synthesis and STxB has a carrier and adjuvant role، also with respect to antibody titers produced، it’s as a vaccine candidate against B. pseudomallei and Shigella dysenteriae.

Abstract: 

Introduction: Pregnancy is an event that changes many views of pregnant woman and it has effect on health، happiness and social roles and sometimes causes psychological problems. The present study aimed to investigate the effect of relaxation training on general health and quality of sleep in pregnant women in the last trimester of pregnancy.Methods: The research design of current study was quasi-experimental and pretest-posttest with control group. A sample size of 32 patients between nulliparous pregnant women who were referred to health care centers in Yazd were selected in the available sampling and randomly divided into two experimental and control groups (Each 16 people). After completing the pre-test, the experimental group was tested for relaxation exercises in seven sessions. Measuring instruments were General Health Questionnaire (Goldberg, 1978), Pittsburgh Sleep Quality Index (Boyce et al., 1989). Data using analysis of co- variance test with SPSS software were analyzed.Results: Results showed that the mean score of the experimental group on general health and quality of sleep was significantly increased compared to control group. That means relaxation training had effect for increasing general health and quality of sleep in pregnant women.Conclusion: The results confirmed the importance and effectiveness of relaxation training for increasing general health and improving sleep quality in pregnant women.

Abstract: 

Introduction: Precocious pubertyin girls refers to onset of puberty before age 8. The purpose of this study therefore was the study of the effect of aerobic training and the use of GnRH agonists on estradiol serum levelsand Gonadotropins in girls with central precocious puberty.Methods: Twenty- five girls with central precocious puberty (aged 7.44±0.34 years) participated in this study. Subjects randomly were divided in to the healthy control group (Without precocious puberty) (n=10) and three experimental groups (drug group) n=10 aerobic exercise and (drug groups) n=8 and aerobic exercise group (n=7). In the experimental group, aerobic program prolonged for 12 weeks 3 days a week 20-60 min a day with 40-80% of Maximum heart rate and additionally GnRH agonists was used as medicine. The estradiol serum levelsand Gonadotropins were measured before, after, and after a follow-up program, respectively. BMI of subjects was measured to assess the effect of the aerobic exercises.Results: Data analysis showed that the aerobic training led to decreased significantly estradiol serum levelsand Gonadotropins in the experimental group (aerobic exercise and aerobic exercise+drug groups) (P≤0/05) The control group had no significant change in the index. BMI had no significant change in the drug group (P=0/06).Conclusions: Considering to the results of this study it could demonstrate that aerobic training and GnRH agonists at the same time can lead to decrease the estradiol serum level sandimprove the Gonadotropins in precocious puberty girls. Aerobic training can also decrease the BMI in girls with central precocious puberty.

Abstract: 

Introduction: Urinary tract infection (UTI) caused by E coli is one of the most common diseases in community. Colonization of E. coli and its attachment to the uroepithelium are mediated by adhesions such as P, Fim and S fimbriae. The present study was aimed to evaluate the prevalence of fimbrial virulence genes in Escherichia coli (E. coli) isolates from the patients with urinary tract infection in Tehran، Boroujerd and Sanandaj cities, Iran.Methods: In this descriptive cross sectional study, 150 clinical isolates of uropathogenic E.coli were collected from the patients with urinary tract infection. All bacterial isolates were identified by standard biochemical laboratory methods; the fim، pap and sfa genes were detected using the PCR and Multiplex-PCR methods.Results: One hundred forty- five (96.66%) isolates were positive for fim gene، one hundred forty (93.33%) isolates were positive for pap gene and seven (4.66) isolates were positive for sfa gene. Whole of the isolates were possessed at least one of the three virulence genes. Six (4%) isolates were positive for all genes.Conclusion: The findings of this study showed the high frequency of fim and P fimbriae among uropathogenic E.coli isolates from the patients with urinary tract infection. Because of the higher prevalence of UTI in the presence of these genes، detection of the genes in urine samples may help in more suspicious and rapid management of UTI.

Abstract: 

Introduction: Nanoparticles (NPs) are one of the antibacterial substances, among them nanoparticles type MgO and Fe2O3 are less toxic to mammalian cells. So, the aim of this study was investigation of combination effects of iron oxide and magnesium oxide nanoparticles on the growth of Staphylococcus aureus and Escherichia coli (E.coli) to achieve the optimum combination of nanoparticles inhibit the growth of Staphylococcus aureus and Escherichia coli in food (juice).Methods: In this experimental research, the effect of MgO and Fe2O3 Nanoparticles compound on Staphylococcus aureus and Escherichia coli bacteria in liquid environment was investigated, and then their effect was investigated separately in juices of carrot, pomegranate and apple via colony count approach. Also, scanning electron microscopy was used to characterize the morphological changes of Staphylococcus aureus and Escherichia coli after antimicrobial treatments. The results of the research were analyzed using one way ANNOVA.Results: The results of the research indicated that in liquid medium, these nanoparticles lead to reduce the growth of both bacteria. compound of 1.5Mg+0.5Fe2O3 was introduced as the most appropriate antibacterial compounds; Staphylococcus aureus sensitivity to Escherichia coli was higher against nanoparticles. The findings of research about the juices revealed that the combined effect of nanoparticles reduced the growth of both bacteria. the combined effect of Fe2o3 and MgO nanoparticles treatments distorted and damaged the cell membrane، resulting in a leakage of intracellular contents and eventually the death of bacterial cells.Conclusion: Nanoparticles in the allowed concentrations have significant effect on Staphylococcus aureus and Escherichia coli bacteria.

Abstract: 

Introduction: Cyclosporine is known as an immunosuppressive drug. Selenium has antioxidant characteristics. This study investigated the synergistic effects of radiation therapy emitted from Gamma rays with the protective effect of selenium after suppressing the immune system of rats with cyclosporine.Methods: 60 adult male mice, randomly divided into 6 groups (5 experimental groups، and one control group). Experimental group number 1: technetium+cyclosporine; group number 2: iodine 131+cyclosporine; group number 3: selenium+cyclosporine; group number 4: cyclosporine+technetium+selenium; group number 5: cyclosporine+iodine131+selenium. Experimental groups 1 and 4 treated with 75 mCi Technetium in a volume of 0.5 ml through intraperitoneal injection. Experimental groups 1 and 5 treated with 0.5 ml iodine 131 (30 mCi). Experimental groups 3, 4 and 5 received 0.1 mg of selenium per kg of their body weight through gavage method. Control group also treated with 0.5 ml normal saline containing 50 mg cyclosporine per kilograms of their body weight to minimize statistical errors. Six hours after treatment with cyclosporine, about 1 ml of heart blood for determining the content of lymphocytes taken and every 6 hours the population of rats has been counted.Results: Percentage of lymphocytes and survival rate was significantly decreased in the experimental group 2. Survival rate of rats in the experimental groups 3, 1, and 4, significantly increased compared to the control group.Conclusion: Treatment with cyclosporine and iodine-131 has a meaningful synergistic effect in the reduction of lymphocyte percentage and also survival time. Selenium and low doses of Technetium has the ability to neutralize the synergistic effect of cyclosporine and iodine-131.
