Abstract

Introduction: Bacteremia is an early problem can lead to life threating conditions like septicemia, shock, and death. According to clinical approach, rapid detection of microorganism in clinical samples is crucial for management of infectious diseases. During the last decades many methods have been developed to rapid diagnosis of infectious agents such as biochemical, immunological, and molecular methods. Among molecular methods, Polymerase Chain Reaction (PCR) is more rapid, specific, and sensitive than conventional methods. Universal primer amplify a conserved region within 16s ribosomal DNA (16s rDNA) sequence that is a constant sequence among all of bacterial species. Detection of bacteria based on 16s rDNA sequence using a universal primer is an accepted method and have been documented in many papers.Methods: In this study, universal primer was used to detection of bacteria in blood specimens obtained from suspected bacteremia patients using PCR technique. During the period between November 2004 and February 2005 one hundred blood specimens collected from Alzahra university hospital in Isfahan, Iran.Results: The results revealed that PCR had 94% specificity and 84% sensitivity comparing to standard blood culture methods.Discussion: According to necessity of rapid initialization of treatment in bacteremia patients clinical diagnosis of bacteremia should be verified by amplification of 16s rDNA by means of a universal primer. This study showed that molecular detection of bacteremia is a fast and applicable method and can be set up in Iranian hospitals.
