THE STUDY OF LINDANE EFFECT ON PENTYLENETERTRAZOLE INDUCED CLONIZURE THRESHOLD IN MICE

Abstract

 Background: The organochlorine insecticide LINDANE, is widely used as a pesticide in agriculture and against ectoparasites in human and veterinary medicine.This drug has been shown to be a seizure- inducing agent in mammals at acute and chronic dosages. The aim of this study was to evaluate the effect of LINDANE on PENTYLENETETRAZOLE- induced seizure threshold and investigate the underlying mechanism(s) responsible of this effect. Materials and Methods: In control group the seizure threshold with PENTYLENETETRAZOLE (PTZ) was 35.29±0.56 mg/kg. Mice were pretreated intraperitoneally with vehicle or LINDANE (5, 10 and 15 mg/kg) before the determination of the PTZ-induced seizure threshold. In another experiment,ivermectin (20, 40 and 60 mg/.kg), c10nazepam (0.1, 0.25 and 0.5 mg/kg) or CARBAMAZEPINE (10, 20 and 40 mg/kg) were injected intraperitoneally in mice alone or in combination with LINDANE (15 mg/kg) before PTZ administration. Results: Results showed that LINDANE significantly decreased seizure threshold. IVERMECTIN, CLONAZEPAM and CARBAMAZEPINE exhibited a dose- dependent increase in the amount of PTZ that was needed to induce a seizure. Pretreatment with IVERMECTIN, c10nazepamor CARBAMAZEPINE on the convulsant property of LINDANE on PTZ- induced seizure threshold showed that all of them significantly reversed the convulsant effect of LINDANE. However, the effects of CLONAZEPAM on PTZ convulsive threshold were more protound. Conclusions: These findings support this hypothesis that an interaction of LINDANE with the GABAA receptor/ er channel is an important mechanism underlying its neurotoxicity and the role of sodium channels in convulsant effect of LINDANE needs more investigation.
